吡格列酮對(duì)糖尿病大鼠異氟烷麻醉后認(rèn)知功能改變的影響

【摘要】:目的:探討吡格列酮對(duì)糖尿病大鼠異氟烷麻醉后認(rèn)知功能改變的影響及可能的機(jī)制方法:選用雄性Sprague Dawley(SD)8個(gè)月齡大鼠52只,,隨機(jī)分出12只為正常對(duì)照組(NC組,n=12)。其余40只大鼠為造模組,均使用一次性腹腔注射50mg/kg鏈脲佐菌素(STZ)誘導(dǎo)糖尿病模型,72小時(shí)后間斷3次取尾靜脈血測(cè)隨機(jī)血糖≥16.7mmol/l為造模成功,NC組大鼠一次性腹腔注射同等量的枸櫞酸鈉緩沖液。造模組40只大鼠全部成功,并隨機(jī)分為糖尿病組(DM組n=20),吡格列酮組(Pio組,n=20)。持續(xù)8周灌胃:正常對(duì)照組和糖尿病組給予相同體積的生理鹽水灌胃,Pio組給予50mg/kg吡格列酮+生理鹽水混懸液灌胃。于灌胃第8周后進(jìn)行Morris水迷宮實(shí)驗(yàn),連續(xù)訓(xùn)練5天,每天4次,記錄逃避潛伏期的均值,第6天予2%異氟烷吸入麻醉2h,第7天完成一次水迷宮測(cè)試,和一次空間探索實(shí)驗(yàn),隨后處死大鼠,進(jìn)行HE染色和免疫組化學(xué)染色法觀察海馬區(qū)椎體神經(jīng)元形態(tài)學(xué)改變和IL-1β,BAX(促凋亡基因)、BCL-2的表達(dá)(抗凋亡基因)。 結(jié)果:DM組大鼠在異氟烷麻醉處理后Morris水迷宮試驗(yàn)逃避潛伏期分別較對(duì)照組和Pio組明顯延長(zhǎng)、在原平臺(tái)象限游泳時(shí)間占總游泳時(shí)間的百分比下降(p<0.05),NC組和Pio組比較沒(méi)有顯著差異;DM組海馬區(qū)炎性因子IL-1β、促凋亡基因BAX表達(dá)上調(diào),抗凋亡基因BCL-2表達(dá)降低及BCL-2/BAX比值下降分別與NC組和Pio組有統(tǒng)計(jì)學(xué)差異(p<0.05),NC組和Pio組比較沒(méi)有統(tǒng)計(jì)學(xué)意義。 結(jié)論:異氟烷吸入麻醉和糖尿病的患病可能會(huì)增加大鼠早期認(rèn)知功能改變發(fā)生的風(fēng)險(xiǎn),其機(jī)制可能是炎癥反應(yīng)的激活;吡格列酮可能具有一定的抗炎效果,并且有可能是通過(guò)該機(jī)制間接的起到神經(jīng)保護(hù)作用,減少糖尿病大鼠異氟烷麻醉后認(rèn)知功能改變的發(fā)生。
