基于PCR-DGGE技術(shù)分析中老年人腸道菌群結(jié)構(gòu)與免疫功能的關(guān)系

【摘要】:目的通過檢測中老年人的各項免疫指標(biāo)和腸道菌群組成,分析中老年人免疫功能與其腸道菌群結(jié)構(gòu)的關(guān)系,探索高齡引起的腸道菌群結(jié)構(gòu)變化對中老年人免疫功能可能的影響。方法選取50~75周歲,健康中老年人99人作為研究對象,抽取空腹靜脈血,檢測CD3~+T細(xì)胞數(shù)、CD4~+T細(xì)胞數(shù)、CD8~+T細(xì)胞數(shù)及血清中的IgA、IgM、IgG。收集各研究對象的糞便,提取糞便細(xì)菌基因組DNA,采用細(xì)菌16S rRNA V6~V8區(qū)通用引物進(jìn)行PCR擴(kuò)增,對擴(kuò)增產(chǎn)物進(jìn)行變性梯度凝膠電泳。采用聚類分析、主成分分析、方差分析等解析研究對象腸道菌群結(jié)構(gòu)特征及與其免疫指標(biāo)的相關(guān)性。結(jié)果研究對象腸道菌群組成表現(xiàn)出高度多態(tài)性和顯著的個體差異,CD3~+T細(xì)胞和CD4~+/CD8~+水平與腸道菌群結(jié)構(gòu)有相關(guān)性,CD4~+/CD8~+偏高的人群與CD4~+/CD8~+偏低和正常人群相比腸道菌群豐富度(S)偏低,但是未觀察到IgA、IgM、IgG與腸道菌群結(jié)構(gòu)存在明顯相關(guān)。結(jié)論中老年人腸道菌群結(jié)構(gòu)與免疫水平相關(guān),腸道菌群多樣性的減少可能會影響其機(jī)體CD8~+T細(xì)胞為中心的非特異性自然免疫功能。
[Abstract]:Objective to analyze the relationship between immune function and intestinal flora structure in middle-aged and elderly people by detecting the immune indexes and composition of intestinal flora, and to explore the possible effects of the changes of intestinal flora structure on immune function in middle-aged and elderly people. Methods 99 healthy adults aged 50 to 75 years were selected as subjects. Fasting venous blood samples were collected to detect the number of CD3 ~ T cells and the number of CD8 ~ T cells in serum. The feces of all the subjects were collected, and the genomic DNA of fecal bacteria was extracted. The amplified products were amplified by PCR using the universal primers of 16s rRNA V6~V8 region, and the amplified products were subjected to denaturing gradient gel electrophoresis (DGGE). Cluster analysis, principal component analysis and variance analysis were used to analyze the structure of intestinal flora and its correlation with immune indexes. Results the composition of intestinal flora showed high polymorphism and significant individual difference. The levels of CD3 ~ T cells and CD4 ~ / CD8 ~ were correlated with intestinal microflora structure. The population with higher CD _ 4 ~ / CD _ 8 ~ was higher than those with lower CD _ 4 ~ / CD _ 8 ~ and normal population. Group richness (S) is low, However, there was no significant correlation between IgA IgMN IgG and intestinal flora structure. Conclusion the structure of intestinal flora is related to the immune level, and the decrease of the diversity of intestinal flora may affect the non-specific natural immune function centered on CD8T cells.
