【摘要】：目的選用ATCC43300標(biāo)準(zhǔn)菌株,建立MRSA感染的大鼠心內(nèi)膜炎模型。方法大鼠經(jīng)右側(cè)頸總動脈插管至左心室,術(shù)后24 h,尾靜脈注射10~3、10~4和10~5 CFU/只感染量的ATCC43300菌液,分別在感染后的1、3、5和7 d處死大鼠,采集心臟贅生物、脾臟和腎臟進(jìn)行菌落計數(shù),同時設(shè)非手術(shù)感染組和手術(shù)非感染組作為對照。結(jié)果感染1 d后,心臟插管大鼠瓣膜處均有贅生物形成,嚴(yán)重者甚至形成壁性贅生物,且隨感染菌濃度的增加,心內(nèi)膜炎感染程度加劇。10~3 CFU/只感染量組、10~4 CFU/只感染量組、10~5 CFU/只感染量組,贅生物細(xì)菌計數(shù)的對數(shù)值分別為7.04±1.38、9.32±1.22、9.78±1.03;感染3 d后,細(xì)菌性心內(nèi)膜炎感染最為嚴(yán)重,細(xì)菌計數(shù)的對數(shù)值最高可達(dá)10.03±0.71;隨后的5~7 d,感染程度隨感染量均有不同程度的下降,10~3 CFU/只感染量組和10~4 CFU/只感染量組,分別下降至6.87±0.63和8.23±1.05;脾臟和腎臟的感染趨勢與心臟大致相同,但感染程度低于心臟。手術(shù)非感染組和非手術(shù)感染組則無贅生物的形成。結(jié)論應(yīng)用該方法建立的模型具有可操作性強(qiáng)、簡單、穩(wěn)定、重復(fù)性好和成功率高的特點(diǎn),更加適用于抗菌藥物機(jī)制、新藥篩選評價及心內(nèi)膜炎相關(guān)性疾病的研究。
[Abstract]:Objective to establish the rat model of endocarditis infected with MRSA by using ATCC43300 standard strain. Methods the rats were intubated to the left ventricle through the right common carotid artery. After 24 hours of operation, the rats were injected into the tail vein with the amount of ATCC43300 bacteria only infected for 10 ~ 3 ~ 10 ~ (4) and 10 ~ (5) CFU/. The rats were killed on the 5th and 7th day after infection, and the cardiac neoplasm, spleen and kidney were collected for colony counting. At the same time, the non-operative infection group and the operative non-infection group were used as the control group. Results after 1 day of infection, there were vegetations in the valve of rats with cardiac catheterization, and even wall vegetations were formed in severe cases, and with the increase of the concentration of infected bacteria. The infection degree of endocarditis was increased. The infection rate of 10 ~ 4 CFU/ only group was 10 ~ 4 CFU/. The logarithmic values of bacterial count of neoplasm were 7.04 鹵1.389.32 鹵1.22 ~ 9.78 鹵1.03.The bacterial endocarditis infection was the most serious after 3 days of infection. The maximum logarithmic value of bacterial count was 10.03 鹵0.71, and the infection degree decreased with the infection amount of 10 ~ 3 CFU/ and 10 ~ 4 CFU/, respectively, to 6.87 鹵0.63 and 8.23 鹵1.05.The infection tendency of spleen and kidney was approximately the same as that of heart. But the degree of infection is lower than that of the heart. No neoplasm was found in the surgical non-infection group and non-operative infection group. Conclusion the model established by this method has the characteristics of strong maneuverability, simplicity, stability, good reproducibility and high success rate, and is more suitable for the study of antimicrobial mechanism, new drug screening and evaluation, and endocarditis related diseases.
