單核增生性李斯特菌hly缺失菌株的構(gòu)建及生物特性的初步鑒定

【摘要】:為研究單核增生性李斯特菌(Listeria monocytogenes,Lm)hly基因?qū)Χ玖Φ挠绊?利用同源重組原理,使用穿梭載體,構(gòu)建單核細(xì)胞增生性李斯特菌野生菌株EGD-e的hly基因缺失菌株EGD-eΔhly。細(xì)菌活性實(shí)驗(yàn)證明缺失菌株生長(zhǎng)狀態(tài)與親本無(wú)差異,但EGD-eΔhly喪失溶血活性,細(xì)胞侵襲能力降低約90%,在10~7 cells/m L腹腔注射條件下不表現(xiàn)動(dòng)物毒性。在轉(zhuǎn)錄水平上EGD-eΔhly的毒力基因inl C、prf A的表達(dá)量分別下降了81%和76%,act A、plc B的表達(dá)量分別提高了2.7倍和1.8倍。hly缺失菌株的成功構(gòu)建及生物特性的初步研究結(jié)果為研究Lm致病機(jī)理提供依據(jù)。
[Abstract]:In order to study the effect of Listeria monocytogenes Lm (Lm) gene on virulence, using the homologous recombination principle and using shuttle vector, the hly gene deletion strain EGD-e 螖 hly. of Listeria monocytogenes EGD-e strain was constructed. The bacterial activity test showed that there was no difference between the growth state of the deficient strain and its parent, but EGD-e 螖 hly lost hemolytic activity, the cell invasion ability decreased about 90%, and no animal toxicity was observed under the condition of 10 7 cells/m L intraperitoneal injection. At the transcriptional level, the expression of virulence gene of EGD-e 螖 hly, inl CfA, decreased by 81% and 76%, respectively, and the expression of act-Aplc B increased by 2.7 times and 1.8 times, respectively, and the results of preliminary study on biological characteristics of Lm-induced strain were as follows: 1. 7 times and 1. 8 times respectively. The mechanism of disease provides basis.
