沙雷菌裂解性噬菌體PS2生物學(xué)特性及其全基因組測(cè)序

【摘要】:目的明確一株粘質(zhì)沙雷菌裂解性噬菌體PS2的生物學(xué)特性及全基因組序列。方法電鏡觀察PS2的形態(tài);雙層平板法分析PS2噬菌譜、生長(zhǎng)曲線及理化穩(wěn)定性;采用MiniBEST Viral RNA/DNA Extraction Kit提取噬菌體基因組,酶解作用鑒定基因組類(lèi)型;構(gòu)建噬菌體PS2測(cè)序文庫(kù),采用MiSeqTM系統(tǒng)測(cè)序,Velvet 1.2.08進(jìn)行序列拼接。結(jié)果 PS2為肌尾型噬菌體。其感染S2菌株的潛伏期為21min,爆發(fā)量為70PFU。PS2在pH5到pH10的環(huán)境中具有較好的穩(wěn)定性,在50℃和60℃的環(huán)境中活性也無(wú)明顯變化。PS2基因組為雙鏈DNA,全長(zhǎng)167 266bp,平均GC含量為41.7%,共預(yù)測(cè)到276個(gè)ORF。BLASTN初步比對(duì)結(jié)果提示其與多株T4-like噬菌體基因組核酸序列具有顯著相似性。結(jié)論分離鑒定了一株粘質(zhì)沙雷菌屬噬菌體,進(jìn)行了生物學(xué)特性、全基因組測(cè)序和生物信息學(xué)初步分析,為噬菌體治療多重耐藥細(xì)菌及噬菌體生物信息學(xué)深入分析奠定了基礎(chǔ)。

Methods the morphology of PS2 was observed by electron microscope, the phage spectrum, growth curve and physicochemical stability of PS2 were analyzed by double-layer plate method, the phage genome was extracted by MiniBEST virus RNA-DNA extraction kit, the genome type was identified by enzymatic hydrolysis, and the phage PS2 sequencing library was constructed. MiSeqTM system was used to sequence Velvet 1.2.08. Results PS2 was a muscular tail phage.
