【摘要】:目的:本研究旨在探討IFN-β和angiostatin分別在離體和在體條件下對(duì)淋巴管內(nèi)皮細(xì)胞生成的影響。 方法:1、淋巴管在體抑制試驗(yàn):觀察活體兔被切斷的淋巴管應(yīng)用抑制因子后的再生情況。2、豬胸導(dǎo)管內(nèi)皮細(xì)胞的培養(yǎng)及形態(tài)學(xué)觀察:應(yīng)用光鏡、同焦顯微鏡、透射電鏡觀察離體胸導(dǎo)管內(nèi)皮細(xì)胞的形態(tài)結(jié)構(gòu)。3、淋巴管內(nèi)皮細(xì)胞離體抑制試驗(yàn):應(yīng)用劃線法和MTT法來(lái)確定IFN-β對(duì)淋巴管內(nèi)皮細(xì)胞生成的抑制作用。4、細(xì)胞調(diào)亡檢測(cè):采用Hoechst法檢測(cè)細(xì)胞凋亡。 結(jié)果:1、手術(shù)切斷兔前或后肢淋巴管5天后,生理鹽水側(cè)(對(duì)照組)的淋巴管已愈合;angiostatin側(cè)(試驗(yàn)組)仍有大量染料滲漏。2、對(duì)培養(yǎng)的淋巴管內(nèi)皮細(xì)胞進(jìn)行了形態(tài)學(xué)觀察。3、劃線法和MTT法均顯示IFN-β對(duì)離體淋巴管內(nèi)皮細(xì)胞的增殖和游走有明顯的抑制作用。4、IFN-β可引起細(xì)胞凋亡 結(jié)論:1、angiostatin對(duì)在體淋巴管的愈合有抑制作用。2、IFN-β對(duì)離體淋巴管內(nèi)皮細(xì)胞的增殖和游走有明顯的抑制作用,且有劑量依賴性。3、IFN-β具有促進(jìn)細(xì)胞凋亡的作用。
[Abstract]:Aim: to investigate the effects of IFN- 尾 and angiostatin on lymphatic endothelial cell formation in vitro and in vivo, respectively. Methods: in vivo inhibition test of lymphatic vessels in vivo: to observe the regeneration of lymphatic vessels in vivo after the application of inhibitory factor in rabbits, and to observe the culture and morphology of endothelial cells in porcine thoracic ducts: using light microscope, isokinetic microscope, Transmission electron microscopy (TEM) was used to observe the morphological structure of thoracic ductal endothelial cells in vitro. The inhibitory effect of IFN- 尾 on lymphatic endothelial cells was determined by the method of underline and MTT, and the apoptosis of lymphatic endothelial cells was detected. Apoptosis was detected by Hoechst method. Results the lymphatic vessels in the saline side (the control group) were healed 5 days after the surgical transection of the lymphatic vessels in the anterior or hind limbs of the rabbits. There was still a large amount of dye leakage in the angiostatin side (experimental group). Morphological observation of cultured lymphatic endothelial cells (LECs) was carried out. The results showed that IFN- 尾 could inhibit the proliferation and migration of lymphatic endothelial cells in vitro. Conclusion: (1) angiostatin can inhibit the healing of lymphatic vessels in vivo. [WT5HZ] IFN- 尾 can inhibit the proliferation and migration of lymphatic endothelial cells in vitro. Moreover, IFN- 尾 can promote apoptosis in a dose-dependent manner.
