【摘要】：目的:通過對動脈粥樣硬化新西蘭兔模型灌胃滌痰湯水煎液,檢驗AS兔的病理組織改變、細(xì)胞、分子生物水平,研究滌痰湯水煎液對AS兔炎性因子的影響。方法:48只雄性新西蘭白兔按隨機(jī)數(shù)字表法,分為6組,每組8只,即空白組、AS模型組、滌痰湯低、中、高劑量組和阿托伐他汀鈣組。12周后,用全自動生化分析儀測定血清中Tc、TG、LDL-c、HDL-c、SOD和MDA的含量;ELISA法測定血清中TNF-α、IL-6、ICAM-1、VCAM-1;于主動脈弓處取直徑3 mm的組織用Western blot檢測作主動脈NF-κB、E-selectin蛋白表達(dá)、用熒光定量PCR檢測TNF-a和IL-6mRNA表達(dá)及用免疫組化法檢測ICAM-1、VCAM-1表達(dá)檢測。結(jié)果:滌痰湯能降低AS兔血清TC、TG、HDL-C、LDL-C、MDA、TNF-α、IL-6、ICAM-1、VCAM-1,增高SOD水平,降低AS兔主動脈NF-κB、E-selectin蛋白的表達(dá),并降低As兔主動脈TNF-α、IL-6 mRNA的表達(dá),且呈劑量依賴性,與模型組比較,差異具有顯著性(P0.01或P0.05)。結(jié)論:動脈粥樣硬化的發(fā)生、發(fā)展與血脂異常、炎癥反應(yīng)的關(guān)系密切;滌痰湯具有調(diào)血脂作用,同時,能提高抗氧化應(yīng)激能力,具有降低致動脈粥樣硬化炎性細(xì)胞因子的作用。
[Abstract]:Objective: to study the effect of Ditan decoction on inflammatory factors of as rabbits by perfusing it into stomach and examining the pathological changes, cellular and molecular biological level of as rabbits. Methods A total of 48 male New Zealand white rabbits were randomly divided into 6 groups with 8 rabbits in each group. The rats were divided into two groups: control group (n = 8), Ditan decoction group (n = 8), low, medium and high dose group and Atto vastatin calcium group (n = 12). The contents of SOD and MDA in serum were determined by automatic biochemistry analyzer. The expression of NF- 魏 BE-selectin in aorta was detected by Western blot, and the expression of TNF- 魏 BE-selectin in aortic arch was detected by Western blot, and the expression of TNF- 偽 IL-6, ICAM-1 and VCAM-1 in serum was determined by enzyme-linked immunosorbent assay (Elisa), and the expression of NF- 魏 BE-selectin in aorta was detected by Western blot. The expression of TNF-a and IL-6 mRNA was detected by fluorescence quantitative PCR and the expression of ICAM-1 and VCAM-1 was detected by immunohistochemistry. Results: Ditan decoction could decrease the level of SOD, decrease the expression of NF- 魏 B and E-selectin and decrease the expression of TNF- 偽 IL-6 mRNA in as rabbit aorta in a dose-dependent manner. The difference was significant compared with the model group (P0.01 or P0.05). Conclusion: the occurrence and development of atherosclerosis are closely related to abnormal blood lipids and inflammatory reaction. Ditan decoction has the effect of regulating blood lipids at the same time it can improve the ability of antioxidant stress and reduce the inflammatory cytokines of atherosclerosis.
