脂肪移植治療DNCB誘導ACD模型小鼠的實驗研究

【摘要】:目的探究脂肪移植治療2,4-二硝基氯苯(DNCB)誘導變應性接觸性皮炎(allergic contact dermatitis,ACD)模型小鼠的效果及其可能機制。方法采用DNCB復制ACD小鼠模型,隨機分成A(脂肪移植治療組)、B(假手術(shù)治療組)、C(藥物治療組)、D組(自然修復組),觀察脂肪移植對ACD小鼠的治療效果。2周后處死小鼠,取背部皮損處全層皮膚組織,蘇木精-伊紅染色法染色觀察各組小鼠皮膚組織病理改變;實時熒光定量聚合酶鏈反應(q RT-PCR)檢測皮損中白介素10(IL-10)、白介素17(IL-17)、干擾素-γ(IFN-γ)的表達。結(jié)果 A、C組可以觀察到表皮厚度恢復正常,角質(zhì)層變薄;B、D組無明顯改變。RT-PCR檢測發(fā)現(xiàn)A、C組皮膚組織中IL-10和IL-17 m RNA水平低于D組,IFN-γm RNA較D組升高(P0.05),而B與D組比較差異無統(tǒng)計學意義(P0.05)。結(jié)論脂肪移植可降低皮損中IL-10和IL-17的表達,促進IFN-γ產(chǎn)生,減輕皮損炎癥。
[Abstract]:Objective to investigate the effect and possible mechanism of fat transplantation in the treatment of allergic contact dermatitis (allergic contact dermatitisus) model mice induced by 2-di-4-dinitrochlorobenzene (DNCBs). Methods A model of ACD mice was established by DNCB. The mice were randomly divided into two groups: group A (fat transplantation group), group B (sham operation group), group D (drug treatment group) and group D (natural repair group). The therapeutic effect of fat transplantation on ACD mice was observed. The histopathological changes of skin were observed by hematoxylin and eosin staining, and the expression of interleukin-10 (IL-10), interleukin-17 (IL-17) and interferon- 緯 (IFN- 緯) were detected by real-time quantitative polymerase chain reaction (Q RT-PCR). Results in group A, the thickness of epidermis returned to normal. RT-PCR analysis showed that the levels of IL-10 and IL-17 mRNA in skin tissue of group A C were lower than those of group D (P0.05), but there was no significant difference between group B and group D (P0.05). Conclusion Fat transplantation can reduce the expression of IL-10 and IL-17, promote the production of IFN- 緯 and alleviate the inflammation of lesions.
