【摘要】:三磷酸腺苷結(jié)合盒轉(zhuǎn)運(yùn)子E1(ABCE1)作為一種高度保守的轉(zhuǎn)運(yùn)蛋白,在生物體的多種細(xì)胞活動(dòng)中發(fā)揮重要作用。ABCE1特異結(jié)合并抑制核糖核酸酶L活性,增強(qiáng)人類免疫缺陷病毒的復(fù)制能力;抑制與細(xì)胞增殖相關(guān)的2-5A/RNase、細(xì)胞質(zhì)L通路;通過(guò)影響蛋白質(zhì)翻譯過(guò)程的起始、終止、核糖體循環(huán)影響真核生物蛋白質(zhì)的合成;穿梭往返于細(xì)胞核質(zhì)之間,可能涉及轉(zhuǎn)錄調(diào)控。將ABCE1的miRNAs或siRNA轉(zhuǎn)入耐藥腫瘤細(xì)胞中抑制其表達(dá),可能提高化療藥物的敏感性,保證腫瘤化療的可行性及可持續(xù)性,為逆轉(zhuǎn)腫瘤耐藥提供新的方法。ABCE1表達(dá)增高或降低均可降低活性氧的活性,因此,ABCE1亦可能成為治療氧化應(yīng)激相關(guān)疾病的新的靶基因。
[Abstract]:Adenosine triphosphate binding cassette transporter E1 (ABCE1), as a highly conserved transporter, plays an important role in many cellular activities. ABCE1 specifically binds and inhibits ribonuclease L activity and enhances the replication ability of human immunodeficiency virus. Inhibition of 2-5A / R Nase, cytoplasmic L pathway associated with cell proliferation; ribosome cycle affects the synthesis of eukaryotic proteins by affecting the initiation and termination of protein translation; shuttling between nucleus and cytoplasm, which may involve transcriptional regulation. Transfection of miRNAs or siRNA of ABCE1 into drug-resistant tumor cells may increase the sensitivity of chemotherapeutic agents and ensure the feasibility and sustainability of tumor chemotherapy. In order to reverse tumor drug resistance, the increased or decreased expression of ABCE1 can decrease the activity of reactive oxygen species, so ABCE1 may also become a new target gene for the treatment of oxidative stress-related diseases.
