【摘要】：目的：研究體外培養(yǎng)的人蛻膜細(xì)胞,經(jīng)不同濃度脂多糖(LPS)作用后,蛻膜細(xì)胞中的炎性因子誘導(dǎo)酶環(huán)氧合酶2(COX-2)表達(dá)差異,探索COX-2過度表達(dá)與蛻膜細(xì)胞感染的相關(guān)性。 方法：收集正常早孕期人工流產(chǎn)術(shù)中的蛻膜組織,進(jìn)行體外原代細(xì)胞培養(yǎng)。當(dāng)蛻膜細(xì)培養(yǎng)3-4代后將其分成5組,分別加入脂多糖使其終濃度分別為Ong/ml、25ng/ml、50ng/ml、100ng/ml、200ng/ml。作用蛻膜細(xì)胞48小時(shí)后,1)通過激光共聚焦-免疫熒光技術(shù)檢測(cè)COX-2蛋白在蛻膜細(xì)胞中的表達(dá)差異2)在收集各組的蛻膜細(xì)胞提取mRNA,并采用半定量逆轉(zhuǎn)錄聚合酶鏈反應(yīng)法(RT-PCR)檢測(cè)各組中COX-2 mRNA的表達(dá)水平,并分析各組間的表達(dá)有無統(tǒng)計(jì)學(xué)差異。 結(jié)果：1)通過激光共聚焦-免疫熒光染色技術(shù)檢測(cè)到各組人蛻膜細(xì)胞后,細(xì)胞質(zhì)及細(xì)胞核中均有COX-2蛋白表達(dá),且加入LPS作用后各實(shí)驗(yàn)組與對(duì)照組相比,COX-2表達(dá)差異顯著,P0.05,有統(tǒng)計(jì)學(xué)意義；且隨著LPS濃度逐漸升高,COX-2蛋白表達(dá)逐漸增強(qiáng)。2)不同濃度LPS作用蛻膜細(xì)胞后,各實(shí)驗(yàn)組與對(duì)照組相比較,COX-2mRNA表達(dá)差異顯著,P0.05,有統(tǒng)計(jì)學(xué)意義；且隨著LPS濃度的增加,COX-2mRNA表達(dá)逐漸增強(qiáng)。 結(jié)論：1)LPS作用于體外培養(yǎng)的人蛻膜細(xì)胞后,COX-2的蛋白表達(dá)增加,且隨著LPS的濃度逐漸增加COX-2蛋白表達(dá)逐漸增強(qiáng)；2)LPS作用于體外培養(yǎng)的人蛻膜細(xì)胞后，，COX-2的mRNA表達(dá)增加,且隨著LPS的濃度逐漸增加COX-2mRNA表達(dá)逐漸增強(qiáng)。
[Abstract]:Aim: to study the difference of expression of inflammatory factor inducible enzyme cyclooxygenase-2 (COX-2) in decidua cells treated with different concentrations of lipopolysaccharide (LPS) in vitro, and to explore the relationship between COX-2 overexpression and decidua cell infection. Methods: the decidual tissue of normal early pregnancy induced abortion was collected and cultured in vitro. When the decidua were cultured for 3 to 4 generations, they were divided into 5 groups, and the final concentration of lipopolysaccharide was 100 ng / ml ~ 100 ng / ml ~ (50) ng 路ml ~ (-1) ~ (-1), respectively. The difference of expression of COX-2 protein in decidual cells was detected by laser confocal immunofluorescence (2) mRNAs were extracted from decidua cells in each group and semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) was used. The expression of COX-2 mRNA in each group was detected by RT-PCR. And analysis of the expression of statistical differences between the groups. Results the expression of COX-2 protein was detected in the cytoplasm and nucleus of decidua cells by laser confocal immunofluorescence staining. Compared with the control group, the expression of COX-2 in each experimental group was significantly different from that in the control group (P 0.05), and the expression of COX-2 protein increased gradually with the increase of LPS concentration. Compared with the control group, the expression of COX-2 mRNA in each experimental group was significantly higher than that in the control group (P 0.05), and increased with the increase of LPS concentration. Conclusion the expression of COX-2 protein in human decidua cells cultured in vitro was increased after treated with LPS, and the expression of COX-2 protein increased gradually with the increase of LPS concentration, and the expression of COX-2 mRNA in human decidual cells cultured in vitro was increased after lipopolysaccharide (LPS) treatment. The expression of COX-2 mRNA increased with the concentration of LPS.
