旋毛蟲與Hela細胞相關(guān)抗原抗體抗腫瘤作用的研究

【摘要】:為了深入研究旋毛蟲抗腫瘤的的作用,本試驗先在體外利用MTT和細胞凋亡的檢測的方法初步確定了旋毛蟲可溶性抗原可以使Hela細胞發(fā)生凋亡,在用ELISA方法初步確定了旋毛蟲與Hela細胞間存在共同抗原,并利用SDS-PAGE和Western-blotting方法測得了相關(guān)抗原的分子量。運用離心超濾裝置及透析等技術(shù)對相關(guān)抗原進行了分離,FITC-免疫組化法對該抗原進行了定位分析。在此基礎(chǔ)上,建立了Hela細胞裸鼠移植瘤動物模型,并對旋毛蟲與Hela細胞相關(guān)抗原抗體進行了抗腫瘤實驗。 實驗結(jié)果顯示:通過MTT法和細胞凋亡實驗證明,旋毛蟲可溶性抗原可以對Hela細胞的增殖具有抑制作用,ELISA法初步證明旋毛蟲與Hela細胞間存在較強的免疫交叉反應(yīng);利用SDS-PAGE和Western-blotting方法獲得了旋毛蟲抗原與Hela細胞相關(guān)抗原的分子量分別是40KDa和55KDa。免疫組化定位分析該相關(guān)抗原位于Hela細胞表面抗原和旋毛蟲的消化道上皮細胞膜上;應(yīng)用相關(guān)抗原抗體在裸鼠體內(nèi)進行抗腫瘤試驗表明,相關(guān)抗原抗體具有抗腫瘤作用,抑瘤率為82.3%,且效果略好于可溶性抗原抗體組。首次以旋毛蟲與Hela細胞相關(guān)抗原抗體為切入點研究旋毛蟲抗腫瘤的機理,為旋毛蟲抗腫瘤機制的研究與開發(fā)提供了理論依據(jù)。
[Abstract]:In order to study the antitumor effect of Trichinella spiralis in vitro, MTT and apoptosis were used to determine that soluble antigen of Trichinella spiralis could induce apoptosis in Hela cells. The common antigen between Trichinella spiralis and Hela cells was preliminarily determined by Elisa, and the molecular weight of the related antigen was measured by SDS-PAGE and Western-blotting. The antigens were isolated by centrifugation ultrafiltration and dialysis. The antigen was localized by FITC- immunohistochemical method. On this basis, an animal model of Hela cell xenografts in nude mice was established, and the antitumor effects of Trichinella spiralis and Hela cell related antigens were tested. The results showed that the soluble antigen of Trichinella spiralis could inhibit the proliferation of Hela cells by MTT assay and apoptosis assay. Elisa showed that there was a strong immune cross reaction between Trichinella spiralis and Hela cells. The molecular weights of Trichinella spiralis antigen and Hela cell associated antigen were 40 KDa and 55 KDa respectively by SDS-PAGE and Western-blotting. Immunohistochemical analysis showed that the associated antigen was located on the surface antigen of Hela cell and the membrane of digestive tract epithelium of Trichinella spiralis. The inhibition rate was 82.3%, and the effect was slightly better than that of soluble antigen antibody group. The antitumor mechanism of Trichinella spiralis was studied by using the antibody against Hela cells for the first time, which provided a theoretical basis for the research and development of antitumor mechanism of Trichinella spiralis.
