大鼠大腦皮質(zhì)星形膠質(zhì)細(xì)胞α2A型腎上腺受體的表達(dá)研究

【摘要】:目的驗(yàn)證大鼠大腦皮質(zhì)星形膠質(zhì)細(xì)胞中的α2A腎上腺素能受體(α2A-AR)的表達(dá)情況,并與α2B腎上腺素能受體(α2B-AR)、α2C腎上腺素能受體(α2C-AR)相比較。方法獲取目的基因α2A-AR、α2B-AR、α2C-AR的RNA反轉(zhuǎn)錄,與內(nèi)參基因β-actin一起進(jìn)行實(shí)時(shí)熒光定量PCR。重復(fù)6組,所得數(shù)據(jù)進(jìn)行統(tǒng)計(jì)學(xué)分析并運(yùn)用2-△△Ct的方法進(jìn)行計(jì)算。結(jié)果大鼠大腦皮質(zhì)星形膠質(zhì)細(xì)胞中確實(shí)表達(dá)了α2A-AR,并且α2A-AR的相對(duì)表達(dá)量是α2B-AR的24.53倍,是α2C-AR的27.98倍。結(jié)論α2A-AR可能為大鼠大腦皮質(zhì)星形膠質(zhì)細(xì)胞接受兒茶酚胺類神經(jīng)遞質(zhì)調(diào)控的主要受體。
[Abstract]:Objective to investigate the expression of 偽 2A adrenergic receptor (偽 2A-ARR) in rat cerebral cortex astrocytes and compare it with 偽 2B adrenoceptor (偽 2B-ARA, 偽 2C adrenoceptor (偽 2C-ARR). Methods RNA reverse transcriptions of target genes 偽 2A-AR, 偽 2B-ARand 偽 2C-AR were obtained, and real-time quantitative PCRs were performed with 尾 -actin. Repeat 6 groups, the data were statistically analyzed and calculated by 2-Ct method. Results 偽 2A-AR was expressed in astrocytes of rat cerebral cortex, and the relative expression of 偽 2A-AR was 24.53 times of 偽 2B-AR and 27.98 times of 偽 2C-AR. Conclusion 偽 2A-AR may be the main receptor of catecholamine neurotransmitters in astrocytes of rat cerebral cortex.
