【摘要】：目的制備并鑒定藍氏賈第鞭毛蟲(簡稱賈第蟲,Giardia lamblia)單克隆抗體(m Ab)。方法以賈第蟲滋養(yǎng)體抗原免疫2只8周齡雌性BALB/c小鼠,利用細胞融合技術建立抗賈第蟲抗原雜交瘤細胞株并制備mAb。制備賈第蟲滋養(yǎng)體可溶性抗原(STA)和排泄-分泌抗原(ESP),采用免疫球蛋白及亞型檢測試劑盒鑒定mAb的類型及亞型。蛋白質印跡(Western blotting)鑒定mAb對賈第蟲滋養(yǎng)體STA和ESP的識別。采用ELISA檢測mAb與賈第蟲滋養(yǎng)體抗原反應性,并檢測與其他寄生蟲抗原的交叉反應。結果獲得12株分泌賈第蟲單克隆抗體細胞株(BB3、CE5、CC10、EG4、GC7、CC1、EF6、DB8、BG10、GH7、HC7和EB2),均為IgG1亞型。其中EB2能識別賈第蟲滋養(yǎng)體STA和ESP中相對分子質量分別為175 000和191 000的蛋白條帶,與大腸埃希菌(Escherichia coli)、豬蛔蟲(Ascaris suum)、人芽囊原蟲(Blastocystis hominis)、日本血吸蟲(Schistosoma japonicum)蟲卵、日本血吸蟲成蟲和衛(wèi)氏并殖吸蟲(Paragonimus westermani)成蟲抗原等均無交叉反應。結論制備的賈第蟲特異性m Ab為Ig G1亞型。
[Abstract]:Objective to prepare and identify Giardia lamblia, a monoclonal antibody against Giardia lamblia. Methods two 8-week-old female BALB / c mice were immunized with Giardia japonica trophozoite antigen. Cell fusion technique was used to establish an anti-Giardia antigen hybridoma cell line and to prepare mAb. Giardia trophozoite soluble antigen (STAA) and excretion-secretory antigen (ESP) were prepared. The type and subtype of mAb were identified by immunoglobulin and subtype detection kit. Western blotting was used to identify the recognition of STA and ESP in Giardia trophozoites by mAb. The reactivity of mAb to Giardia trophozoite antigen and cross reaction with other parasitic antigens were detected by Elisa. Results Twelve McAb cell lines (BB3, CE5, CC10, EG4, GC7, CC1, EF6, BG10, GH7, HC7 and EB2) were obtained, all of which belonged to IgG1 subtype. EB2 could identify the protein bands with molecular weight of 175 000 and 191 000 in giardia trophozoite STA and ESP, respectively, with Escherichia coli, Ascaris suumium, Blastocystis hominisus, Schistosoma japonicum, Schistosoma japonicum. There was no cross reaction between adult Schistosoma japonicum and Paragonimus westermanii. Conclusion the specific mAb of Giardia was Ig G 1 subtype.
