慢性復(fù)合應(yīng)激對家兔多個(gè)系統(tǒng)功能的影響及Orcu-DRB等位基因的測定

【摘要】:目的:本文通過對慢性復(fù)合型應(yīng)激狀態(tài)下家兔的體重、生存時(shí)間、血清糖皮質(zhì)激素、生化指標(biāo)水平、抗雞卵核抗體和總抗氧化能力等指標(biāo)的觀察,探討慢性復(fù)合型應(yīng)激對家兔多個(gè)系統(tǒng)功能狀態(tài)的影響。 方法:由束縛、浸水、隔離限制三種應(yīng)激方式共同組成而建立本次慢性復(fù)合型應(yīng)激動物模型,并從以下三個(gè)方面來研究慢性復(fù)合型應(yīng)激對家兔產(chǎn)生的影響。 1、在實(shí)施慢性復(fù)合型應(yīng)激的過程中,記錄家兔每日體重變化、存活數(shù)與死亡數(shù),通過分析慢性復(fù)合型應(yīng)激后較應(yīng)激前家兔的體重變化,并在后期進(jìn)行生存分析,統(tǒng)計(jì)累積生存率,分析應(yīng)激時(shí)間與家兔累積生存率的關(guān)系,并由此對本實(shí)驗(yàn)動物模型的建立進(jìn)行評價(jià)。 2、通過檢測慢性復(fù)合應(yīng)激前后家兔血清中總蛋白、白蛋白、葡萄糖、谷丙轉(zhuǎn)氨酶、肌酐及乳酸脫氫酶等常見臨床生化指標(biāo)以及糖皮質(zhì)激素、抗雞卵核抗體及總抗氧化能力的水平,研究慢性復(fù)合型應(yīng)激對上述各項(xiàng)指標(biāo)的影響。 3、取用家兔耳緣動脈血,提取全血基因組DNA,,采用PCR-SSP技術(shù)檢測以O(shè)rcu-DRB*02型等位基因?yàn)榇淼耐肈RB等位基因,并對家兔攜帶與未攜帶Orcu-DRB*02型等位基因者糖皮質(zhì)激素及體重變化進(jìn)行了初步的分析。 結(jié)果: 1、通過觀察分析應(yīng)激后較應(yīng)激前家兔的體重變化,發(fā)現(xiàn)應(yīng)激后家兔的體重顯著性下降,并于應(yīng)激第31天達(dá)到最低點(diǎn)。在應(yīng)激第4天時(shí),應(yīng)激組家兔發(fā)生第一例死亡。在應(yīng)激第15天時(shí),累積生存率為50%,并且家兔累計(jì)生存率在應(yīng)激后呈持續(xù)顯著性下降趨勢。 2、應(yīng)激后尤其在第四周家兔血清中的葡萄糖較應(yīng)激前相比顯著下降,應(yīng)激后的血清總抗氧化能力較應(yīng)激前相比降低;應(yīng)激后家兔體內(nèi)的糖皮質(zhì)激素水平較應(yīng)激前有顯著增高。而應(yīng)激后血清中總蛋白、球蛋白、白蛋白、谷丙轉(zhuǎn)氨酶、谷草轉(zhuǎn)氨酶、GGT、肌酐、乳酸脫氫酶、抗雞卵核抗體與應(yīng)激前相比沒有顯著性差異。 3、Orcu-DRB等位基因檢測結(jié)果:36只家兔中,Orcu-DRB*02型等位基因陽性表達(dá)的家兔有3只,陽性率為8.33%。未攜帶Orcu-DRB*02型等位基因的家兔體重隨著應(yīng)激時(shí)間的增加而有上升的趨勢,而攜帶Orcu-DRB*02型等位基因的家兔體重隨著應(yīng)激時(shí)間的增加而有下降的趨勢。 結(jié)論: 1.慢性復(fù)合應(yīng)激后家兔的體重增長明顯受抑,說明此次建立動物模型的方法有效。 2.慢性復(fù)合應(yīng)激后家兔血清中的葡萄糖較應(yīng)激前相比有顯著下降;應(yīng)激后的血清總抗氧化能力較應(yīng)激前相比降低,說明葡萄糖、總抗氧化能力這兩項(xiàng)指標(biāo)對慢性應(yīng)激比較敏感。 3.在新西蘭兔中可以檢測出Orcu-DRB*02型等位基因。
[Abstract]:Objective: to observe the body weight, survival time, serum glucocorticoid, biochemical index, anti-oocyte antibody and total antioxidant ability of rabbits under chronic complex stress. To investigate the effects of chronic complex stress on the functional states of multiple systems in rabbits. Methods: the animal model of chronic compound stress was established by three stress modes: restraint, immersion and isolation. The effects of chronic compound stress on rabbits were studied from the following three aspects. 1. In the course of implementing chronic compound stress, the changes of daily weight, the number of survival and the number of deaths in rabbits were recorded. The changes of weight of rabbits after chronic compound stress were analyzed compared with those before stress, and survival analysis was carried out at the later stage. The relationship between the stress time and the cumulative survival rate of rabbits was analyzed, and the establishment of the animal model was evaluated. 2. Serum total protein, albumin, glucose, alanine aminotransferase, creatinine, lactate dehydrogenase and glucocorticoid in serum of rabbits before and after chronic multiple stress were detected. To study the effect of chronic complex stress on the above indexes in the level of anti-oocyte antibody and total antioxidant capacity. 3. The genomic DNA was extracted from rabbit auricular artery blood, and the DRB alleles represented by Orcu-DRB*02 type allele were detected by PCR-SSP technique. The changes of glucocorticoid and body weight in rabbits with and without Orcu-DRB*02 alleles were analyzed. Results: 1. By observing and analyzing the weight changes of rabbits after stress, it was found that the weight of rabbits decreased significantly after stress, and reached the lowest point on the 31st day of stress. On the fourth day of stress, the first death occurred in the stress group. On the 15th day of stress, the cumulative survival rate was 50%, and the cumulative survival rate of rabbits continued to decrease significantly after stress. (2) after stress, especially in the fourth week, the serum glucose decreased significantly, and the total antioxidation capacity of serum decreased after stress, and the level of glucocorticoid in rabbits after stress was significantly higher than that before stress. However, there was no significant difference in serum total protein, globulin, albumin, alanine aminotransferase, glutamic oxaloacetic transaminase (GGTs), creatinine, lactate dehydrogenase and anti-oocyte antibody. 3The results of Orcu-DRB allele analysis showed that there were 3 rabbits with positive expression of Orcu-DRBF02 allele in 36 rabbits, and the positive rate was 8.33%. The weight of rabbits without Orcu-DRB*02 allele increased with the increase of stress time, while the weight of rabbits with Orcu-DRB*02 allele decreased with the increase of stress time. Conclusion: 1. The weight gain of rabbits was significantly inhibited after chronic multiple stress, which indicated that the method of establishing animal model was effective. 2. The total antioxidant capacity of serum after stress was lower than that before stress, which indicated that glucose and total antioxidant capacity were sensitive to chronic stress. 3. The Orcu-DRB*02 allele can be detected in New Zealand rabbits.
