龜羚帕安丸對(duì)帕金森病大鼠模型中腦黑質(zhì)細(xì)胞Nestin蛋白,Nestin mRNA及DAT蛋白的影響

【摘要】:目的:觀察龜羚帕安丸對(duì)帕金森病(PD)大鼠黑質(zhì)細(xì)胞巢蛋白(Nestin),多巴胺轉(zhuǎn)運(yùn)體(DAT)及Nestin mRNA表達(dá)的影響。方法:大鼠黑質(zhì)致密部注射6-羥基多巴胺(6-OHDA),后大鼠ip阿撲嗎啡(APO)誘導(dǎo)旋轉(zhuǎn)建立PD大鼠模型。PD大鼠隨機(jī)分為模型組,多巴絲肼組,龜羚帕安丸高、中、低劑量組,同時(shí)設(shè)立假手術(shù)組,模型組。假手術(shù)組每日ig同等容積的生理鹽水;多巴絲肼組ig多巴絲肼混懸液(10 mg·kg~(-1)·d~(-1));龜羚帕安丸高、中、低劑量組ig龜羚帕安丸混懸液(4,2,1 g·kg~(-1)·d~(-1)),連續(xù)30 d。免疫組化法檢測(cè)各組大鼠中腦黑質(zhì)Nestin,DAT蛋白表達(dá),原位雜交法檢測(cè)各組大鼠中腦黑質(zhì)Nestin mRNA的表達(dá)。結(jié)果:模型組黑質(zhì)細(xì)胞Nestin,DAT蛋白,Nestin mRNA表達(dá)均顯著低于假手術(shù)組(P0.01);各治療組Nestin蛋白,Nestin mRNA,DAT蛋白表達(dá)均顯著高于模型組(P0.01)。結(jié)論:龜羚帕安丸治療帕金森病的作用機(jī)制可能與上調(diào)Nestin,DAT表達(dá),促進(jìn)神經(jīng)的修復(fù)和再生,增加多巴胺(DA)含量有關(guān)。
[Abstract]:Aim: to observe the effect of Guilingpaan pill on the expression of nestin, dopamine transporter (DATT) and Nestin mRNA in substantia nigra of rats with Parkinson's disease (PD). Methods: PD rat model was established by injecting 6-hydroxydopamine (6-OHDAA) into the substantia nigra compactly and then by IP apomorphine (APO). PD rats were randomly divided into three groups: model group, dobutazide group, high, middle and low dose group of Guilingpaan pills. At the same time, set up sham operation group, model group. The sham operation group was given the same volume of normal saline daily; the dobutazid group was given intravenously with dobutazid suspension of 10 mg kg 路kg ~ (-1) 路d ~ (-1) ~ (-1) d ~ (-1); the group of Guilingpaan pills was high, medium and low dose group, and the group of ig was given 4 ~ 2g / kg / kg ~ (-1) 路d ~ (-1) 路d ~ (-1) ~ (-1) of the suspension of Guilingpaan Pill (ig) for 30 days. The expression of Nestin mRNA in substantia nigra was detected by immunohistochemistry and in situ hybridization was used to detect the expression of Nestin mRNA in substantia nigra. Results: the expression of nestin mRNA in substantia nigra cells in the model group was significantly lower than that in the sham-operated group, and the expression of nestin mRNA-DAT protein in all treatment groups was significantly higher than that in the model group. Conclusion: the mechanism of Guilingpaan Pill in the treatment of Parkinson's disease may be related to the up-regulation of Nestinine DAT expression, the promotion of nerve repair and regeneration, and the increase of dopamine (DA) content.
