【摘要】：目的通過高脂飼養(yǎng)GK大鼠建立一種符合人類糖尿病周圍神經(jīng)病變(DPN)發(fā)病特點(diǎn),又操作簡單的DPN大鼠模型。方法 7~8周齡SPF級雄性自發(fā)性2型糖尿病GK大鼠30只,喂養(yǎng)高脂飼料造模。另取30只SPF級正常Wistar大鼠作為正常組,喂養(yǎng)普通飼料。每周監(jiān)測動物血糖、體質(zhì)量、飲水量、飼料量。分別于干預(yù)8周、12周、16周后檢測血清糖化血紅蛋白、坐骨神經(jīng)傳導(dǎo)速度,取對側(cè)坐骨神經(jīng)做HE染色,TUNEL染色計(jì)算細(xì)胞凋亡指數(shù)。結(jié)果隨著造模時(shí)間延長,GK大鼠逐漸出現(xiàn)多飲、多食、生長遲緩等表現(xiàn)。與正常組相比,造模12周及16周的大鼠血糖、糖化血紅蛋白升高(P0.01),感覺神經(jīng)傳導(dǎo)速度降低(P0.01),運(yùn)動神經(jīng)傳導(dǎo)速度有一定下降趨勢(12周P0.05,16周P0.05),坐骨神經(jīng)病理形態(tài)及雪旺細(xì)胞凋亡情況均提示DPN的形成,并與正常大鼠形成鮮明對照(凋亡指數(shù)P0.01)。結(jié)論長期高脂飼料喂養(yǎng)的GK大鼠是DPN研究的良好模型,12周是較為成熟且經(jīng)濟(jì)的造模時(shí)間。
[Abstract]:Objective to establish a DPN rat model with high fat feeding in accordance with the characteristics of diabetic peripheral neuropathy (DPN). Methods Thirty SPF grade male GK rats with spontaneous type 2 diabetes were fed with high fat diet. Another 30 normal Wistar rats of SPF grade were taken as normal group and fed with normal diet. Blood sugar, body mass, drinking water and feed quantity were monitored weekly. Serum glycosylated hemoglobin and sciatic nerve conduction velocity were measured after 8 weeks and 12 weeks and 16 weeks, respectively. The contralateral sciatic nerves were taken for HE staining and Tunel staining to calculate apoptosis index. Results with the prolongation of model making time, GK rats gradually appeared more drink, more food and growth retardation. Compared with the normal group, the blood glucose levels of the rats at 12 and 16 weeks after modeling were compared with those in the control group. The HbA1c increased, the sensory nerve conduction velocity decreased, the motor nerve conduction velocity decreased, and the motor nerve conduction velocity decreased. The histopathological morphology of sciatic nerve and the apoptosis of Schwann cells indicated the formation of DPN. In contrast to normal rats, the apoptotic index (P0.01) was significantly different from that in normal rats. Conclusion GK rats fed with high fat diet for a long time are a good model for DPN study. 12 weeks is a more mature and economical time to model GK rats.
