【摘要】：目的研究銀屑1號對咪喹莫特誘導小鼠銀屑病模型血清中炎性因子IL-6,IL-17,INF-γ和角質(zhì)形成細胞內(nèi)核轉錄因子-κB(NF-κB)及NF-κB mRNA表達的影響。方法入選動物隨機分為5組:正常組(Normal)、模型組(Model)、銀屑1號高(HD)、中(MD)、低(LD)劑量組,除正常組外,所有小鼠外涂咪喹莫特軟膏14日誘導為銀屑病模型,銀屑1號高、中、低劑量組給藥劑量分別為50g/(kg·d),25g/(kg·d),12.5g/(kg·d),連續(xù)10d,正常組及模型組予等量生理鹽水對照。檢測表皮垂直厚度,血液蛋白芯片法檢測血清I-L6,IL-17,INF-γ水平,蘇木精-依紅染色觀察皮損組織結構改變,免疫組化法檢測NF-κB含量,應用逆轉錄-聚合酶鏈反應檢測NF-κB mRNA表達情況。結果與正常組比較,模型組角質(zhì)增厚,鏡下T淋巴細胞浸潤明顯,其炎性因子、NF-κB蛋白及基因表達均升高(P0.01);同模型組比較,銀屑1號高、中劑量組IL-6,IL-17,INF-γ水平下降明顯(P0.01);高、中、低劑量組表皮增厚程度明顯降低(P0.01),NF-κB蛋白水平明顯降低(P0.01),中、低劑量組NF-κB mRNA表達降低(P0.05)。結論銀屑1號對銀屑病小鼠模型的角質(zhì)增殖和炎性反應具有抑制作用,抑制NF-κB蛋白及基因的過度表達是其作用機制之一。
[Abstract]:Objective to study the effects of Yinshi 1 on the expression of IL-6, IL-17, INF- 緯, NF- 魏 B, NF- 魏 B, and NF- 魏 B mRNA in the serum of mice psoriasis model induced by Imiquimod. Methods the animals were randomly divided into 5 groups: normal control group, model group, silver chip 1 high HDD group, middle MDD group, low LDD group. All mice were treated with Imiquimod ointment for 14 days as psoriasis model, silver chip 1 was high and medium. In the low dose group, the dosage was 25g / kg / kg 50g/(kg / L / kg / kg DX for 10 days, respectively. The normal group and the model group were given the same amount of normal saline control. The vertical thickness of epidermis, the level of serum I-L6, IL-17, INF- 緯, the changes of tissue structure, the content of NF- 魏 B and the expression of NF- 魏 B mRNA were detected by hematoxylin staining, immunohistochemistry method and reverse transcription-polymerase chain reaction respectively. Results compared with the normal group, the keratinocytes of the model group were thicker, the infiltration of T lymphocytes was obvious under microscope, and the expression of the inflammatory factor NF- 魏 B and its gene were all increased in the model group, compared with the model group, the level of IL-6, IL-17, INF- 緯 in the silver chip 1 group was higher than that in the middle dose group, and the level of IL-6, IL-17, INF- 緯 in the model group was significantly lower than that in the model group. In low dose group, the level of NF- 魏 B protein was significantly decreased, and the expression of NF- 魏 B mRNA was decreased in low dose group. Conclusion YS-1 has inhibitory effect on keratinocyte proliferation and inflammatory response in psoriatic mice, and inhibiting the overexpression of NF- 魏 B protein and gene is one of its mechanisms.
