慢性胃炎病癥結(jié)合模型大鼠與能量代謝的實(shí)驗(yàn)研究

【摘要】:目的: 1、探討胃炎大鼠脾胃虛證、濕熱證模型的造模方法 2、探討不同證型胃炎大鼠在能量代謝方面的指標(biāo)差異 材料和方法: 選取40只清潔級(jí)SD大鼠,隨機(jī)分成四組,脾胃濕熱組,脾胃虛弱組,模型組,正常組。造模期間觀察大鼠的毛色、食量、體重的變化。造模四周后,濕熱組8只,脾胃虛弱組9只,模型組、正常組各10只。取胃黏膜和骨骼肌通過(guò)酶促反應(yīng)和定磷方法,檢測(cè)各個(gè)組別能量代謝指標(biāo)ATP酶活性,比較相關(guān)性。取胃竇部光鏡下通過(guò)HE染色,分析病理結(jié)果。 結(jié)果: 骨骼肌和胃黏膜ATP酶活性測(cè)試表明:脾胃濕熱組與正常組相比,P0.05,有統(tǒng)計(jì)學(xué)差異。脾胃虛弱組與正常組相比,P0.05,有統(tǒng)計(jì)學(xué)差異。模型組與正常組相比,P0.05,有統(tǒng)計(jì)學(xué)差異。 胃竇部HE病理染色顯示:正常組:大鼠黏膜上皮細(xì)胞排列整齊,腺體形態(tài)規(guī)則,大小一致,固有層偶見(jiàn)炎性細(xì)胞浸潤(rùn)。模空組:胃黏膜血管擴(kuò)張充血,腺體形態(tài)尚規(guī)則,但固有層可見(jiàn)少量炎癥細(xì)胞浸潤(rùn)。提示已有慢性胃炎的病理學(xué)改變。濕熱組:胃黏膜缺損,脫落,壞死,固有層見(jiàn)大量炎癥細(xì)胞浸潤(rùn)。脾虛組:胃黏膜缺損,脫落,壞死,固有層可見(jiàn)炎癥細(xì)胞較濕熱少一些。 結(jié)論: 脾胃虛弱組數(shù)值明顯偏低,表明處于長(zhǎng)期處于脾胃虛弱狀態(tài)的大鼠ATP合成減少,機(jī)體能量代謝減少。脾胃濕熱組數(shù)值明顯偏高,表明基礎(chǔ)酶活力和能量代謝增高。胃竇部病理染色顯示:各組炎癥程度,脾胃是熱組脾胃虛弱組模型組正常組。
[Abstract]:Objective: 1. To explore the method of making model of spleen and stomach deficiency syndrome and damp-heat syndrome in rats with gastritis. 2. To explore the difference of energy metabolism in rats with different types of gastritis. Materials and methods: Forty SD rats of clean grade were randomly divided into four groups: spleen and stomach damp-heat group, spleen and stomach weakness group, model group and normal group. The changes of color, food intake and body weight of the rats were observed during modeling. Four weeks later, there were 8 rats in damp-heat group, 9 in spleen and stomach weakness group, 10 in model group and 10 in normal group. Gastric mucosa and skeletal muscle were measured by enzymatic reaction and phosphorus fixation to detect the activity of ATP enzyme in each group of energy metabolism indexes. The pathological results of gastric antrum were analyzed by HE staining under light microscope. Results: The activity of ATP enzyme in skeletal muscle and gastric mucosa showed that there was significant difference between the spleen and stomach dampness heat group and the normal group (P 0.05). Compared with the normal group, the spleen and stomach weak group had significant difference (P 0.05). There was statistical difference between model group and normal group (P 0.05). The HE staining of gastric antrum showed that in normal group, the epithelial cells were arranged neatly, the glands were regular, the size was the same, and inflammatory cell infiltration was occasionally seen in the lamina propria. Model empty group: gastric mucosal vasodilation hyperemia, glandular shape is regular, but there are a few inflammatory cells infiltration in lamina propria. It suggests that there are pathological changes in chronic gastritis. Damp-heat group: gastric mucosal defect, abscission, necrosis, lamina propria a large number of inflammatory cell infiltration. Spleen deficiency group: gastric mucosal defect, shedding, necrosis, inflammatory cells visible in the lamina propria less than damp and heat. Conclusion: The value of spleen and stomach weakness group was significantly lower, which indicated that ATP synthesis and energy metabolism were decreased in rats with chronic weakness of spleen and stomach. The value of spleen and stomach damp-heat group was significantly higher than that of the control group, indicating that the activity of basic enzymes and energy metabolism were increased. The pathological staining of gastric antrum showed that the degree of inflammation in each group and spleen and stomach were normal group.
