肉蓯蓉顆粒劑對帕金森病大鼠模型黑質(zhì)紋狀體多巴胺能神經(jīng)元的保護作用研究

【摘要】:目的:探討肉蓯蓉顆粒劑對帕金森病大鼠模型黑質(zhì)紋狀體多巴胺能神經(jīng)元的保護作用。方法:采用6-羥基多巴胺單側(cè)雙靶點顱內(nèi)定向注射法制備帕金森病大鼠模型,3周后據(jù)阿樸嗎啡誘導(dǎo)的不自主運動,篩選造模成功大鼠,隨機分為空白組,模型組,假手術(shù)組,肉蓯蓉顆粒劑低劑量組(1 g·kg-1·d-1)和肉蓯蓉顆粒劑高劑量組(3 g·kg-1·d-1),灌胃相應(yīng)藥物共4周。記錄阿樸嗎啡誘導(dǎo)的大鼠向健側(cè)旋轉(zhuǎn)的圈數(shù);采用免疫組化研究觀察黑質(zhì)內(nèi)酪氨酸羥化酶(TH)的表達;和免疫印跡法檢測(Western-blot)觀察紋狀體內(nèi)α-突出核蛋白(α-synuclein)、酪氨酸羥化酶(TH)、多巴胺轉(zhuǎn)運蛋白(DAT)的表達;高效液相色譜-質(zhì)譜聯(lián)用(HPLC-MS/MS)法檢測定大鼠紋狀體多巴胺(DA)、3,4-二羥基苯乙酸(DOPAC)、高香草酸(HVA)的含量。結(jié)果:用藥4周后,肉蓯蓉組向健側(cè)旋轉(zhuǎn)的圈數(shù)與模型組比較明顯減少(P0.05),黑質(zhì)內(nèi)TH陽性細胞較模型組明顯增多,紋狀體內(nèi)α-synuclein表達下調(diào)、TH、DAT表達上調(diào)。紋狀體DA、DOPAC含量明顯升高(P0.05或P0.01)。高劑量組效果優(yōu)于低劑量組。結(jié)論:肉蓯蓉顆粒對帕金森病大鼠模型黑質(zhì)和紋狀體的多巴胺能神經(jīng)元具有神經(jīng)保護作用。
[Abstract]:Aim: to investigate the protective effect of Cistanche deserticola granules on dopaminergic neurons in substantia nigra striatum of Parkinson's disease rats. Methods: a rat model of Parkinson's disease was established by intracranial injection of 6-hydroxydopamine (6-hydroxydopamine) at one side and two targets. After 3 weeks' involuntary exercise induced by apomorphine, successful rats were selected and randomly divided into three groups: blank group, model group and sham operation group. Cistanche deserticola granules low dose group (1 g kg-1 d-1) and Cistanche granule high dose group (3 g kg-1 d -1) were gavaged for 4 weeks. The number of cycles of contralateral rotation induced by apomorphine was recorded, and the expression of tyrosine hydroxylase (THH) in substantia nigra was observed by immunohistochemistry. The expression of 偽 -synuclein (tyrosine hydroxylase) in striatum was detected by Western blot. High performance liquid chromatography-mass spectrometry (HPLC-MS / MS) method was used to determine the content of dopamine Dachlorine 3, 4-dihydroxyphenylacetic acid (DOPA) in the striatum of rats. Results: after 4 weeks of administration, the number of rotation circles in the contralateral direction of Cistanche deserticola group was significantly lower than that in the model group, the number of th positive cells in the substantia nigra was significantly higher than that in the model group, and the expression of 偽 -synuclein was down-regulated in the striatum. The content of DOPAC in striatum was significantly higher than that in control group (P 0.05 or P 0.01). The effect of high dose group was better than that of low dose group. Conclusion: Cistanche deserticola granule has neuroprotective effect on dopaminergic neurons in substantia nigra and striatum of Parkinson's disease rats.
