誘導(dǎo)腫瘤細(xì)胞HeLa為多能性干細(xì)胞的研究

【摘要】:將攜帶4種轉(zhuǎn)錄因子的逆轉(zhuǎn)錄病毒共同感染HeLa細(xì)胞,感染后的細(xì)胞鋪到滋養(yǎng)層細(xì)胞MEF上培養(yǎng),細(xì)胞長(zhǎng)出類似胚胎干細(xì)胞的克隆團(tuán),這種克隆團(tuán)可以擴(kuò)增,并進(jìn)一步誘導(dǎo)分化為神經(jīng)元,同時(shí)免疫熒光檢測(cè)細(xì)胞分化不同天數(shù)后Pax6,Sox1和βⅢ-tubulin的表達(dá).結(jié)果表明,腫瘤細(xì)胞HeLa在Oct4、Sox2、c-Myc、Klf4等4種轉(zhuǎn)錄因子的作用下成功重編程為多能干細(xì)胞,同時(shí)可進(jìn)一步分化為神經(jīng)元.
