【摘要】：研究桂枝茯苓丸逆轉(zhuǎn)人卵巢癌耐順鉑細(xì)胞株(SKOV3/DDP)腫瘤模型的耐藥作用及其對(duì)荷瘤BALB/c裸鼠生存率的影響,探討其可能的作用機(jī)制。采用Cell Counting Kit-8試劑盒檢測(cè)SKOV3/DDP對(duì)順鉑和紫杉醇的耐藥性;采用SKOV3/DDP細(xì)胞建立卵巢癌耐藥腫瘤模型,模型BALB/c裸鼠隨機(jī)分為桂枝茯苓丸濃縮液16 g·kg~(-1)·d~(-1)(高)、8 g·kg~(-1)·d~(-1)(中)、4g·kg~(-1)·d~(-1)(低)劑量(中藥組)、順鉑/紫杉醇組(化療組)、低劑量桂枝茯苓丸與化療聯(lián)用組(中西聯(lián)用組)和空白對(duì)照組,檢測(cè)各處理組的抑瘤率。觀察各組BALB/c裸鼠的生存情況;并采用熒光定量PCR檢測(cè)各組腫瘤組織中MDR1 mRNA的相對(duì)表達(dá)。結(jié)果是SKOV3/DDP細(xì)胞對(duì)順鉑和紫杉醇的耐藥倍數(shù)分別為4.13倍和3.85倍。隨著桂枝茯苓丸濃縮液劑量的增加,抑瘤作用增強(qiáng),裸鼠生存率提高,腫瘤組織MDR1 mRNA表達(dá)相對(duì)于對(duì)照組逐漸下降。西藥化療組在耐藥模型裸鼠中的抑瘤效果高于中藥組,中西藥聯(lián)用后抑瘤效果顯著高于西藥化療組;與其他各組相比,西藥化療組的裸鼠平均體重明顯下降,差異均具有顯著性(P0.05,P0.01)。中西聯(lián)用組裸鼠生存率高于桂枝茯苓丸低劑量組和西藥化療組。西藥化療組MDR1mRNA表達(dá)相對(duì)于對(duì)照組顯著增加,中西藥聯(lián)用組MDR1 mRNA表達(dá)相對(duì)于對(duì)照組顯著下降,差異均具有顯著性(P0.05,P0.01)。說(shuō)明桂枝茯苓丸能夠逆轉(zhuǎn)SKOV3/DDP耐藥性卵巢癌模型裸鼠的耐藥性,提高裸鼠生存率,其機(jī)制可能與抑制MDR1 mRNA表達(dá)有關(guān)。
[Abstract]:To study the effect of Guizhi Fuling pill on reversing the drug-resistance of human ovarian cancer cell line SKOV3 / DDP and its effect on the survival rate of nude mice bearing tumor BALB/c, and to explore its possible mechanism. The drug resistance of SKOV3/DDP to cisplatin and paclitaxel was detected by Cell Counting Kit-8 kit, and the drug-resistant tumor model of ovarian cancer was established by SKOV3/DDP cells. Model BALB/c nude mice were randomly divided into two groups: Guizhi Fuling Pill concentrated liquid (16g / kg ~ (-1) ~ (1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) (Chinese medicine group), Cisplatin / paclitaxel group (chemotherapy group), low dose Guizhi Fuling Pill combined with chemotherapy group (combined with Chinese and western medicine) and blank control group (P < 0.05), and the model mice were divided into three groups: Cisplatin / paclitaxel group (chemotherapy group) and blank control group. The tumor inhibition rate of each treatment group was detected. The survival of each group of BALB/c nude mice was observed, and the relative expression of MDR1 mRNA in tumor tissues was detected by fluorescence quantitative PCR. The results showed that the resistance of SKOV3/DDP cells to cisplatin and paclitaxel was 4.13 and 3.85 times respectively. With the increase of the concentration of Guizhi Fuling Pill, the anti-tumor effect was enhanced, the survival rate of nude mice was increased, and the expression of MDR1 mRNA in tumor tissue was gradually decreased compared with the control group. The anti-tumor effect of the western medicine chemotherapy group was higher than that of the traditional Chinese medicine group, and that of the western medicine chemotherapy group was significantly higher than that of the western medicine chemotherapy group, and compared with other groups, the average weight of the nude mice in the western medicine chemotherapy group was significantly lower than that in the western medicine chemotherapy group. The difference was significant (P 0.05, P 0.01). The survival rate of nude mice in combination group was higher than that of Guizhi Fuling pill group and western medicine chemotherapy group. Compared with the control group, the expression of MDR1mRNA in the western medicine chemotherapy group was significantly higher than that in the control group, and the expression of MDR1 mRNA in the combination of Chinese and western medicine group was significantly lower than that in the control group. The results indicated that Guizhi Fuling pills could reverse the drug resistance of SKOV3/DDP resistant ovarian cancer model and improve the survival rate of nude mice. The mechanism of Guizhi Fuling pills might be related to the inhibition of MDR1 mRNA expression.
