睡眠剝奪對大鼠學(xué)習(xí)記憶和神經(jīng)遞質(zhì)的影響及中西藥的干預(yù)作用

【摘要】:目的: 1.研究快速眼動睡眠剝奪(RSD)和對氯苯丙氨酸(PCPA)所致完全性失眠兩種睡眠剝奪(SD)模型對大鼠學(xué)習(xí)記憶及血漿中單胺類神經(jīng)遞質(zhì)去甲腎上腺素(NE)、多巴胺(DA)及5-羥色胺(5-HT)的含量的影響。 2.研究松郁安神方和阿普唑侖對兩種SD模型干預(yù)后大鼠學(xué)習(xí)記憶及血漿中單胺類神經(jīng)遞質(zhì)含量的變化。 方法: 健康雄性Sprague-Dawley大鼠88只,體重200±10g,隨機(jī)選取其中40只用于建立SD模型。采用改良多平臺法(MMPM)建立RSD模型,采用腹腔注射PCPA建立完全性失眠模型,分成5組:空白對照(CC)組、大平臺環(huán)境對照(TC)組、快速眼動睡眠剝奪(RSD)組、腹腔注射PCPA組、腹腔注射生理鹽水(NS)對照組。另外48只用于藥物干預(yù):建立兩種SD模型后分別用松郁安神方和阿普唑侖灌胃5天。大鼠分成6組:NS+蒸餾水組、PCPA+蒸餾水組、RSD+中藥組、RSD+西藥組、PCPA+中藥組、PCPA+西藥組。各組動物實驗前后行Morris水迷宮測試大鼠的學(xué)習(xí)記憶能力。實驗結(jié)束后,腹主動脈取血,用高效液相色譜-電化學(xué)檢測法(HPLC-ECD)檢測血漿中NE、DA、5-HT的含量。 結(jié)果: 1.大鼠學(xué)習(xí)記憶能力變化: 造模后Morris水迷宮測試各組的潛伏期較造模前均下降(P0.05)。RSD造模后,RSD組的潛伏期比TC組、CC組明顯延長(P0.05)。藥物干預(yù)后,與RSD組相比,RSD+中藥組和RSD+西藥組大鼠潛伏期縮短(P0.05)。 2.血漿中NE、DA、5-HT含量的變化: (1)SD模型組:與CC組、TC組、NS組相比,RSD組、PCPA組NE含量升高(P0.05)。與CC組、PCPA組相比,RSD組DA含量下降,與NS組相比,PCPA組DA含量升高(P0.05)。與CC組、NS組比較,RSD、PCPA組5-HT含量下降(P0.05)。 (2)藥物干預(yù)組:與RSD組、PCPA組、RSD+中藥組相比,PCPA+中藥組、PCPA+西藥組NE含量下降;與RSD組比較,RSD+西藥組NE含量下降;與PCPA組、PCPA+蒸餾水組比較,PCPA+中藥組DA下降;與PCPA組、RSD組比較,RSD+西藥組、PCPA+西藥組、PCPA+中藥組5-HT含量升高。 結(jié)論: 1.RSD使大鼠的學(xué)習(xí)記憶能力受損。PCPA對學(xué)習(xí)記憶能力無顯著影響。中藥及西藥灌胃均可使RSD大鼠學(xué)習(xí)記憶得到改善,提示藥物干預(yù)能提高機(jī)體的學(xué)習(xí)記憶能力。 2.RSD使血漿中NE含量升高,DA、5-HT含量下降,PCPA失眠使血漿中NE、DA含量上升,5-HT含量下降。 3.中藥干預(yù)后RSD大鼠血漿中5-HT含量升高,PCPA失眠大鼠NE、DA含量下降,5-HT含量升高。西藥干預(yù)后RSD大鼠NE含量下降、5-HT含量上升, PCPA失眠大鼠5-HT含量上升。神經(jīng)遞質(zhì)含量的變化提示藥物干預(yù)可能通過改變失眠者體內(nèi)的神經(jīng)遞質(zhì)調(diào)節(jié)睡眠。
[Abstract]:Objective: 1. The effects of rapid eye movement sleep deprivation (RSDs) and complete insomnia induced by p-chlorphenylalanine (PCPA) on learning and memory and the contents of monoamine neurotransmitter norepinephrine (NE), dopamine (DA) and serotonin (5-HT) in plasma of rats were studied. 2. To study the changes of learning and memory and the content of monoamine neurotransmitters in plasma after the intervention of Songyu Anshen recipe and alprazolam on two SD models in rats. Methods: 88 healthy male Sprague-Dawley rats, weighing 200 鹵10 g, were randomly selected to establish SD model. The RSD model was established by modified multi-platform method and the complete insomnia model was established by intraperitoneal injection of PCPA. The model was divided into five groups: blank control group, large platform environment control group, rapid eye movement sleep deprivation group, and intraperitoneal injection of PCPA group. The control group was treated by intraperitoneal injection of NS. The other 48 rats were used for drug intervention: two SD models were established and treated with Songyu Anshen recipe and alprazolam respectively for 5 days. Rats were divided into 6 groups: PCPA distilled water group and PCPA distilled water group. The learning and memory abilities of rats were tested by Morris water maze before and after the experiment. After the experiment, the blood was taken from the abdominal aorta and the plasma NEDAD 5-HT was detected by high performance liquid chromatography-electrochemical detection (HPLC-ECD). Results: 1. Changes of learning and memory ability in rats: The latency of Morris water labyrinth test group was significantly longer than that of TC group. After drug intervention, compared with RSD group, the latency of rats in RSD group and RSD group was shortened (P 0.05). 2. Changes of 5-HT in plasma: The content of NE in PCPA group was higher than that in TC group (CC group) and NS group (P 0.05). Compared with CC group, the DA content of PCPA group was lower than that of PCPA group, and the DA content of PCPA group was higher than that of NS group. Compared with CC group and NS group, the 5-HT content of RSDN PCPA group was decreased (P 0.05). (2) Drug intervention group: compared with RSD group, the content of NE in PCPA group was lower than that in RSD group, the NE content in western medicine group was lower than that in RSD group, and the DA level in PCPA group was lower than that in PCPA group. Compared with the PCPA group, the content of 5-HT in the PCPA group was higher than that in the western medicine group. Conclusion: 1.RSD had no significant effect on learning and memory ability. Both Chinese medicine and western medicine could improve the learning and memory of RSD rats, suggesting that drug intervention could improve the ability of learning and memory. The content of NE in plasma was increased by 2.RSD and the content of 5-HT in plasma was decreased. The content of NEDA in plasma was increased and the content of 5-HT in plasma was decreased. 3. The content of 5-HT in plasma of RSD rats was increased after the intervention of traditional Chinese medicine. The content of NEDA in rats with RSD insomnia decreased and the content of 5-HT increased. After intervention of western medicine, NE content decreased and 5 HT content increased in RSD rats, and 5-HT content increased in PCPA insomnia rats. Changes in neurotransmitters suggest that drug intervention may regulate sleep by altering neurotransmitters in insomnia patients.
