左歸丸聯(lián)合溫和灸對骨質(zhì)疏松癥模型大鼠骨密度、骨保護(hù)素mRNA及核因子κB受體活化因子配體mRNA的影響

【摘要】:目的探討左歸丸聯(lián)合溫和灸治療骨質(zhì)疏松癥的可能作用機(jī)制。方法 40只雌性SD大鼠隨機(jī)分為空白組、模型組、左歸丸組、左歸丸+溫和灸組,每組10只。模型組、左歸丸組、左歸丸+溫和灸組采用手術(shù)完整摘除雙側(cè)卵巢法制造骨質(zhì)疏松癥模型。造模3個(gè)月后左歸丸組予左歸丸10 ml/(kg·d)灌胃,左歸丸+溫和灸組予左歸丸10 ml/(kg·d)灌胃+長強(qiáng)穴艾灸治療,空白組和模型組予生理鹽水10 ml/(kg·d)灌胃。各組均干預(yù)6周。干預(yù)前后測定大鼠股骨近端及第2腰椎骨密度,干預(yù)后通過PCR法檢測大鼠骨組織骨保護(hù)素(OPG)mRNA和核因子κB受體活化因子配體(RANKL)mRNA表達(dá)水平。結(jié)果與空白組比較,模型組股骨、腰椎骨密度,骨組織OPG mRNA表達(dá)及OPG/RANKL降低,RANKL mRNA表達(dá)上升(P0.05或P0.01);與模型組比較,左歸丸組、左歸丸+溫和灸組大鼠股骨、腰椎骨密度,骨組織OPG mRNA表達(dá)及OPG/RANKL均升高,RANKL mRNA表達(dá)降低(P0.05或P0.01),且左歸丸+溫和灸組大鼠股骨、腰椎骨密度,骨組織OPG mRNA表達(dá)及OPG/RANKL均較左歸丸組升高,RANKL mRNA表達(dá)較左歸丸組降低(P0.05)。結(jié)論左歸丸聯(lián)合溫和灸能明顯升高骨質(zhì)疏松癥大鼠的骨密度,上調(diào)OPG mRNA表達(dá)、下調(diào)RANKL mRNA表達(dá)可能是其作用機(jī)制之一。
[Abstract]:Objective to explore the possible mechanism of Zuo Gui Pill combined with mild moxibustion in the treatment of osteoporosis. Methods Forty female Sprague-Dawley rats were randomly divided into blank group, model group, Zuo Gui Wan group and Zuo Gui Wan mild moxibustion group with 10 rats in each group. Model group, Zuo Gui Wan group and Zuo Gui Wan mild moxibustion group were treated with complete ovariectomy to make osteoporosis model. After 3 months, Zuo Gui Pill group was given Zuo Gui Pill (10 ml/(kg d), Zuo Gui Wan mild moxibustion Group (ZJW 10 ml/(kg) was treated with Changqiang Point moxibustion, and the blank group and model group were treated with normal saline for 10 ml/(kg d). All groups were treated for 6 weeks. Bone mineral density (BMD) of proximal femur and 2 lumbar vertebrae were measured before and after intervention. After intervention, the expression of osteoprotegerin (OPG) mRNA and nuclear factor- 魏 B receptor activator ligand (RANKLN) mRNA were detected by PCR method. Results compared with the blank group, the bone mineral density (BMD) of femur and lumbar vertebrae, the expression of OPG mRNA in bone tissue and the decrease of mRNA expression of RANKL in the model group were increased (P0.05 or P0.01), compared with the model group, the bone mineral density of femur and lumbar vertebrae in the model group, Zuo Gui Wan mild moxibustion group, and Zuo Gui Wan mild moxibustion group were increased. The expression of OPG mRNA and OPG/RANKL in bone tissue were increased. The expression of RANKL mRNA was decreased (P 0.05 or P 0.01). The bone mineral density (BMD), bone tissue OPG mRNA expression and OPG/RANKL expression in the mild moxibustion group were higher than those in the Zuo Gui Wan group. The expression of RANKL mRNA was lower than that of the left Gui Wan group (P 0.05). Conclusion Zuo Gui Pill combined with mild moxibustion can significantly increase bone mineral density (BMD), upregulate OPG mRNA expression and down-regulate RANKL mRNA expression in osteoporosis rats.
