【摘要】：鐵是維持細(xì)胞生長(zhǎng)繁殖及代謝活動(dòng)所必需的元素之一,機(jī)體內(nèi)絕大多數(shù)游離鐵是由轉(zhuǎn)鐵蛋白運(yùn)輸傳遞。轉(zhuǎn)鐵蛋白(Tf)是血漿中主要的結(jié)合并轉(zhuǎn)運(yùn)鐵的β球蛋白,主要負(fù)責(zé)運(yùn)載由胃腸道吸收的鐵和由紅細(xì)胞降解釋放的鐵。Tf的金屬離子結(jié)合位點(diǎn)由2個(gè)酪氨酸、1個(gè)天冬氨酸和1個(gè)組氨酸殘基構(gòu)成。Tf除了轉(zhuǎn)運(yùn)鐵外同時(shí)還具有許多生理生化功能,如抗菌活性、促細(xì)胞生長(zhǎng)分化及細(xì)胞保護(hù)等,是一類多功能蛋白。此外,研究發(fā)現(xiàn)Tf還被發(fā)現(xiàn)可作為一種新型的藥物主動(dòng)靶向載體,在腫瘤靶向治療領(lǐng)域中具有廣闊的應(yīng)用前景。該文從Tf的結(jié)構(gòu)特性、多樣性、生物學(xué)功能及臨床應(yīng)用等方面對(duì)Tf進(jìn)行系統(tǒng)性綜述。
[Abstract]:Iron is one of the essential elements to maintain cell growth, reproduction and metabolism. Most of the free iron in the body is transported by transferrin. Transferrin (TF) is the major binding and transporting 尾 globulin in plasma. The metal ion-binding sites mainly responsible for carrying iron absorbed by the gastrointestinal tract and iron. TF released by red blood cells are composed of two tyrosine, one aspartic acid and one histidine residue. Multiple physiological and biochemical functions, It is a kind of multifunctional protein, such as antibacterial activity, promoting cell growth and differentiation, cell protection and so on. In addition, TF has been found to be a novel active drug targeting carrier and has a broad application prospect in the field of tumor targeting therapy. This paper reviews the structure, diversity, biological function and clinical application of TF.
