血小板低下對(duì)樹(shù)突狀細(xì)胞在體遷移與歸巢模式的影響研究

【摘要】:目的:探討血小板低下對(duì)過(guò)繼性樹(shù)突狀細(xì)胞(DC)在體內(nèi)遷移模式的影響。方法:腹腔注射抗CD41單克隆抗體以建立血小板低下小鼠模型。將帶報(bào)告基因的骨髓來(lái)源DC經(jīng)腳墊和靜脈兩種途徑輸注至血小板低下和正常小鼠體內(nèi),用活體成像技術(shù)監(jiān)測(cè)DC在小鼠體內(nèi)的遷移和組織內(nèi)分布情況。結(jié)果:經(jīng)抗GPⅡb抗體處理組小鼠體內(nèi)80%以上血小板被清除。DC經(jīng)腳墊回輸72 h后,在血小板低下組遷移到乆窩淋巴結(jié)(PLN)和腹股溝淋巴結(jié)(ILN)的比例分別為(0.32±0.02)%和(0.02±0.01)%;對(duì)照組上述兩部位內(nèi)為(0.27±0.15)%和(0.02±0.02)%。兩組之間無(wú)統(tǒng)計(jì)學(xué)差異(P0.05)。經(jīng)靜脈輸注DC 72 h后,血小板低下組和對(duì)照組小鼠組織內(nèi)的分布均主要集中在脾臟、肝臟引流淋巴結(jié)、肺臟和肝臟等,且分布比例無(wú)統(tǒng)計(jì)學(xué)差異。結(jié)論:小鼠體內(nèi)血小板低下至不足20%時(shí),對(duì)經(jīng)皮下和靜脈兩種途徑輸注的DC在體內(nèi)遷移和組織分布無(wú)明顯影響。
[Abstract]:Aim: to investigate the effect of thrombocytopenia on the migration pattern of adoptive dendritic cells (DC) in vivo. Methods: mouse model of thrombocytopenia was established by intraperitoneal injection of anti-CD41 monoclonal antibody. DC derived from bone marrow with reporter gene was infused into mice with thrombocytopenia and normal mice via foot pad and vein.
