【摘要】：目的采用免疫組化(IHC)方法觀察日本血吸蟲感染破壞小鼠脾臟淋巴濾泡結(jié)構(gòu)的現(xiàn)象。方法日本血吸蟲感染小鼠(20條尾蚴/鼠)8周后剖殺,取脾臟樣本石蠟包埋、切片,先以蘇木素-伊紅(HE)染色觀察小鼠脾臟淋巴濾泡整體結(jié)構(gòu),再以IHC法用抗B220、CD21及Ki67抗體標(biāo)記淋巴濾泡內(nèi)B細(xì)胞、濾泡樹突狀細(xì)胞(FDC)及生發(fā)中心細(xì)胞,觀察濾泡內(nèi)具體細(xì)胞分布。結(jié)果 HE染色顯示,感染日本血吸蟲小鼠脾臟淋巴濾泡結(jié)構(gòu)模糊難辨,相較于正常小鼠濾泡數(shù)目及面積均顯著減少;IHC染色發(fā)現(xiàn),日本血吸蟲感染小鼠脾臟T、B淋巴細(xì)胞分區(qū)及FDC網(wǎng)絡(luò)結(jié)構(gòu)和生發(fā)中心結(jié)構(gòu)均消失。結(jié)論日本血吸蟲感染可顯著破壞小鼠脾臟淋巴濾泡結(jié)構(gòu)。
[Abstract]:Objective to observe the effect of Schistosoma japonicum infection on the structure of splenic lymphatic follicles in mice by immunohistochemical staining (IHC). Methods Twenty cercariae / mice infected by Schistosoma japonicum were dissected after 8 weeks. The spleen samples were embedded in paraffin and sectioned. The whole structure of lymphoid follicles was observed by hematoxylin and eosin (HEH) staining. B cells in lymphoid follicles, follicular dendritic cells (DC) and germinal center cells were labeled with anti-B220 CD21 and Ki67 antibodies by IHC method. The distribution of specific cells in follicles was observed. Results HE staining showed that the structure of splenic lymphatic follicles in Schistosoma japonicum infected mice was fuzzy and difficult to distinguish. Compared with normal mice, the number and area of follicles were significantly decreased by IHC staining. In mice infected with Schistosoma japonicum, the distribution of T _ (B) lymphocytes, the network structure of FDC and the structure of germinal center disappeared. Conclusion Schistosoma japonicum infection can significantly destroy the lymphatic follicular structure in mice.
