3種流感病毒檢測方法的比較

【摘要】:目的對病毒分離培養(yǎng)法、實時熒光PCR法和雞胚培養(yǎng)法3種檢測流感病毒的方法和結果進行比較研究,篩選快速準確的檢測方法,為流感監(jiān)測與防控工作提供及時可靠的病原學依據。方法同時采用病毒分離培養(yǎng)法、實時熒光PCR法和雞胚培養(yǎng)法對2014年10月~2015年3月牡丹江市流感監(jiān)測哨點醫(yī)院采集的流感樣病例咽拭子樣本進行檢測,比較檢測結果。結果實時熒光PCR法的流感病毒核酸檢測陽性率為15.52%,其中169份為季節(jié)性H3N2流感病毒,73份為B型Yamagata株;病毒分離培養(yǎng)法的陽性率為6.29%,分離出63株季節(jié)性H3N2流感病毒,35株B型Yamagata株;雞胚分離培養(yǎng)法檢出率為0.64%。3種方法中,實時熒光PCR法的陽性率最高,病毒分離培養(yǎng)法次之,雞胚分離培養(yǎng)法陽性率最低。結論實時熒光PCR法比病毒分離培養(yǎng)法和雞胚培養(yǎng)法靈敏快捷、特異性強、敏感度高,適用于流感病毒的快速檢測。
[Abstract]:Objective to compare the three methods and results of virus isolation and culture, real-time fluorescence PCR and chicken embryo culture, and to screen rapid and accurate methods for detection of influenza virus. To provide timely and reliable etiological basis for influenza surveillance and prevention and control. Methods simultaneously, virus isolation culture method, real-time fluorescence PCR method and chicken embryo culture method were used to detect the throat swabs of influenza cases collected from Mudanjiang influenza surveillance sentinel hospital from October 2014 to March 2015, and the results were compared. Results the positive rate of influenza virus nucleic acid detected by real-time fluorescent PCR method was 15.52, of which 169 were seasonal H3N2 influenza virus and 73 were type B Yamagata strain, and the positive rate of virus isolation culture method was 6.29 and 63 seasonal H3N2 influenza virus type B Yamagata strains were isolated. The positive rate of chicken embryo isolation culture method was 0.64.3, the positive rate of real-time fluorescence PCR method was the highest, virus isolation culture method was the second, and chicken embryo isolation culture method had the lowest positive rate. Conclusion the method of real-time fluorescence PCR is more sensitive, specific and sensitive than virus isolation culture and chicken embryo culture. It is suitable for rapid detection of influenza virus.
