【摘要】：目的建立半抗原2,4-二硝基氟苯(DNFB)誘發(fā)C57BL/6小鼠變應(yīng)性接觸性皮炎(ACD)模型,檢測(cè)瘙癢介質(zhì)胃泌素釋放肽(GRP),P物質(zhì)和TRPV-1分子在背根神經(jīng)節(jié)(DRG)中的表達(dá)并探討它們對(duì)在ACD伴發(fā)的慢性瘙癢的作用。方法外用DNFB建立C57BL/6小鼠的ACD模型,觀察皮炎癥狀和皮膚病理等指標(biāo),檢測(cè)血清總IgE和IL-4濃度,并分析GRP,P物質(zhì)和TRPV-1在頸部背根神經(jīng)節(jié)(DRG)的表達(dá)水平變化。結(jié)果與對(duì)照組相比,DNFB模型組小鼠頸背部皮膚表現(xiàn)出明顯炎癥性皮損,病理提示表皮增厚、真皮肥大細(xì)胞浸潤(rùn),血清IgE和IL-4明顯升高,GRP,P物質(zhì)和TRPV-1的mRNA在頸部DRG表達(dá)升高。結(jié)論外用半抗原DNFB成功建立了C57BL/6小鼠ACD模型,GRP,P物質(zhì)和TRPV-1在ACD模型瘙癢中有重要作用。
[Abstract]:Objective to establish an ACD-induced allergic contact dermatitis (ACD) model in C57BL/6 mice induced by hapten 2N 4-dinitrofluorobenzene (DNFB). To detect the expression of pruritus mediators gastrin releasing peptide (gastrin releasing peptide) and TRPV-1 in DRG and to explore their effects on chronic pruritus associated with ACD. Methods the ACD model of C57BL/6 mice was established by DNFB. The symptoms of dermatitis and skin pathology were observed. The serum total IgE and IL-4 levels were measured. The expression of GRPP-P and TRPV-1 in the cervical dorsal root ganglion (DRG) was analyzed. Results compared with the control group, the skin of the mice in the DNFB group showed obvious inflammatory lesions. The pathological results showed that the epidermis thickened, the dermis mast cells infiltrated, and the serum IgE and IL-4 increased significantly the expression of GRP P and mRNA of TRPV-1 in the neck DRG. Conclusion the ACD model of C57BL/6 mice was successfully established by external use of hapten DNFB and TRPV-1 and GRP P played an important role in the pruritus of ACD model.
