【摘要】:研究下調(diào)CDH17基因?qū)δ强啥∧退幍娜宋赴㎝GC-803細(xì)胞裸鼠移植瘤模型的影響,為臨床上尋求治療胃癌新方案提供理論依據(jù)。以人胃癌MGC-803細(xì)胞作為研究材料,建立該細(xì)胞的裸鼠移植瘤模型。同時(shí),用HE染色法觀察瘤體組織病理學(xué)形態(tài)。結(jié)果顯示,RT-PCR和Western blotting實(shí)驗(yàn)中siCDH17慢病毒對(duì)胃癌MGC-803細(xì)胞中CDH17的轉(zhuǎn)錄水平和蛋白質(zhì)表達(dá)水平均具有干擾效果(P0.05)。隨后將siCDH17慢病毒干擾的人胃癌MGC-803細(xì)胞以及對(duì)照細(xì)胞進(jìn)行裸鼠皮下移植瘤實(shí)驗(yàn)。移植后第6天各組均見皮下腫瘤結(jié)塊,采用那可丁藥物處理并實(shí)時(shí)檢測(cè)腫瘤生長(zhǎng)情況。結(jié)果發(fā)現(xiàn),siCDH17組腫瘤體積和質(zhì)量明顯小于對(duì)照組和siNC組(P0.05)。且HE染色也發(fā)現(xiàn)與對(duì)照組比較,siCDH17組的瘤體腫瘤細(xì)胞生長(zhǎng)受到抑制。因此,下調(diào)CDH17基因可以增加胃癌MGC-803細(xì)胞對(duì)那可丁的敏感性,抑制胃癌MGC-803細(xì)胞的生長(zhǎng),最終阻滯裸鼠移植瘤的生長(zhǎng)。
[Abstract]:To study the effect of down-regulation of CDH17 gene on the xenograft tumor model of human gastric cancer MGC-803 cells in nude mice with drug resistance of nakodin, and to provide a theoretical basis for seeking a new regimen for the treatment of gastric cancer in clinic.The xenograft tumor model of human gastric cancer MGC-803 cells was established in nude mice.At the same time, the histopathological morphology of the tumor was observed by HE staining.The results showed that siCDH17 lentivirus in RT-PCR and Western blotting could interfere with the transcription and protein expression of CDH17 in MGC-803 cells of gastric cancer.Then human gastric cancer MGC-803 cells and control cells interfered with siCDH17 lentivirus were subcutaneously transplanted into nude mice.On the 6th day after transplantation, subcutaneous tumor caking was observed in all groups.The results showed that the tumor volume and mass in siCDH17 group were significantly lower than those in control group and siNC group (P 0.05).He staining also showed that tumor cell growth of siCDH 17 group was inhibited compared with control group.Therefore, down-regulation of CDH17 gene can increase the sensitivity of gastric cancer MGC-803 cells to nakodin, inhibit the growth of gastric cancer MGC-803 cells, and eventually block the growth of xenografts in nude mice.
