SGT基因雜合缺失小鼠血液生理生化指標(biāo)分析

【摘要】:目的分析SGT基因雜合缺失對不同年齡小鼠血液指標(biāo)和代謝的影響。方法分別采集10周齡、26周齡SGT基因雜合缺失(SGT~(+/-))小鼠和野生型小鼠眼眶血液,用全自動血細(xì)胞分析儀和生化分析儀檢測常用血液生理指標(biāo)和生化指標(biāo),進(jìn)行不同年齡和基因型間統(tǒng)計比較。結(jié)果與同基因型低齡小鼠對比,高齡小鼠多項血液生理生化指標(biāo)均發(fā)生增齡性改變。與同齡野生型對照相比,10周齡SGT~(+/-)小鼠大血小板比率降低,未結(jié)合型膽紅素濃度降低(P0.05),其余指標(biāo)均未見統(tǒng)計學(xué)差異;26周齡SGT~(+/-)小鼠血小板計數(shù)、血小板比容和中性粒細(xì)胞計數(shù)降低,堿性磷酸酶、未結(jié)合型膽紅素、膽固醇及甘油三酯等指標(biāo)改變有統(tǒng)計學(xué)意義(P0.05)。結(jié)論SGT基因雜合缺失能夠誘發(fā)高齡小鼠部分血液、血清生化指標(biāo)的改變。
[Abstract]:Objective to analyze the effect of SGT gene heterozygosity on blood index and metabolism in mice of different ages.Methods Orbital blood samples were collected from 10-week and 26-week-old SGT gene heterozygous deletion mice and wild-type mice respectively. The normal blood physiological and biochemical indexes were detected by automatic blood cell analyzer and biochemical analyzer.Statistical comparison was made between different ages and genotypes.Results compared with the young mice with the same genotype, aging changes occurred in many blood physiological and biochemical indexes of the aged mice.Compared with the wild-type control of the same age, the ratio of large platelets and the concentration of unconjugated bilirubin decreased in 10-week-old SGT (/ -) mice.The changes of platelet volume and neutrophil count, alkaline phosphatase, unconjugated bilirubin, cholesterol and triglyceride were statistically significant (P 0.05).Conclusion the loss of heterozygosity of SGT gene can induce the changes of blood and serum biochemical indexes in aged mice.
