貫穿縫扎聯(lián)合打孔法構(gòu)建兔耳難愈性創(chuàng)面模型

【摘要】:背景:以往制作的兔耳難愈性創(chuàng)面模型僅局限于單純的缺血模型,缺血淤血模型少見(jiàn)。 目的:建立一種方法簡(jiǎn)單、效果確實(shí)的兔耳難愈性創(chuàng)面模型。 方法:健康新西蘭大白兔20只即兔耳40個(gè),隨機(jī)分為縫扎+打孔組(實(shí)驗(yàn)組)和單純打孔組(對(duì)照組)兩組,每組兔耳20個(gè)。實(shí)驗(yàn)組貫穿兔耳根部背腹側(cè)皮膚和軟骨,間斷縫扎中央、頭側(cè)所有動(dòng)、靜脈,縫扎后第10天在耳背側(cè)用活檢打孔器制作3個(gè)深達(dá)軟骨面、直徑6mm的圓形創(chuàng)面。對(duì)照組在正常兔耳(血管未行縫扎)背側(cè)用活檢打孔器制作與實(shí)驗(yàn)組完全相同的3個(gè)圓形創(chuàng)面。兔耳縫扎、打孔創(chuàng)面每日外涂0.5%碘伏3次。觀察兔耳大體情況、創(chuàng)面愈合時(shí)間,打孔后第7、14、21天行蘇木精-伊紅(hematoxylin-eosin, HE)染色及血管內(nèi)皮細(xì)胞生長(zhǎng)因子(vascular endothelial growth factor, VEGF)免疫組織化學(xué)染色,分別觀察創(chuàng)面肉芽組織生長(zhǎng)、表皮化程度及肉芽組織中血管內(nèi)皮細(xì)胞生長(zhǎng)因子的表達(dá)情況。 結(jié)果:實(shí)驗(yàn)組兔耳縫扎后立即呈現(xiàn)紫黑色,第10、20、30天為淡紫黑色、紫色、淡紫色。實(shí)驗(yàn)組、對(duì)照組創(chuàng)基肉芽組織生長(zhǎng)、表皮化時(shí)間分別為第3-4天、第16-18天和第1-2天、第11-13天。實(shí)驗(yàn)組與對(duì)照組比較,在創(chuàng)面愈合時(shí)間(P0.01)、創(chuàng)面肉芽組織生長(zhǎng)、表皮化時(shí)間均延遲,血管內(nèi)皮細(xì)胞生長(zhǎng)因子表達(dá)實(shí)驗(yàn)組低于對(duì)照組(P0.01)。 結(jié)論:貫穿縫扎聯(lián)合打孔法制作的兔耳難愈性創(chuàng)面模型方法簡(jiǎn)單、效果確實(shí)可靠,可作為缺血淤血因素所致難愈性創(chuàng)面修復(fù)的實(shí)驗(yàn)研究模型。
[Abstract]:Background: Previous wound healing of rabbit ear model making limited to simple models of ischemia, a rare model of ischemia and blood stasis.
Objective: to establish a simple method of wound of rabbit ears, refractory model does effect.
Methods: 20 healthy New Zealand white rabbits: 40 rabbits, randomly divided into suture and punch group (experimental group) and simple perforation group (control group) two groups, each group of 20 rabbits. The experimental group through rabbit ear root dorsal ventral skin and cartilage, interrupted suture central, all side head move, vein, suture after tenth days in the ear side with biopsy punch 3 deep cartilage 6mm diameter circular wounds. In the control group of normal rabbit ears (without vascular suture) 3 circular dorsal wound biopsy punch fabrication and experimental group rabbit exactly the same. Ear suture, 3 punch wound daily coated with 0.5% iodophor. Rabbit ears to observe the general situation, the wound healing time, perforation after 7,14,21 days of hematoxylin eosin staining (hematoxylin-eosin, HE) and vascular endothelial growth factor (vascular endothelial, growth factor, VEGF) immunohistochemical staining were observed in the granulation group The growth factor, the degree of epidermis and the expression of vascular endothelial growth factor in the granulation tissue.
Results: the experimental group rabbit ear suture violet black immediately after 10,20,30 days, the purple black, purple, pale purple. The experimental group, the control group was the growth of granulation tissue and epidermal time respectively for the first 3-4 days, the 16-18 day and the 1-2 day, the 11-13 day. The experimental group compared with control group and in wound healing time (P0.01), the growth of granulation tissue and epidermis of time delay, the expression of vascular endothelial growth factor in experimental group than in the control group (P0.01).
Conclusion: the suture of wound healing in rabbit ear model ligation combined punch made of simple, reliable effect, can be used as an experimental model for the study of repair caused by ischemia and congestion factors of refractory wounds.
