佐劑提高過敏反應(yīng)動(dòng)物模型敏感性研究

【摘要】:目的:通過觀察茵梔黃注射液和生脈注射液佐劑組與未聯(lián)用佐劑組在過敏反應(yīng)發(fā)生時(shí)的敏感指標(biāo)的含量變化,研究此類佐劑提高BN大鼠過敏反應(yīng)動(dòng)物模型穩(wěn)定性和靈敏度的能力。方法:將弗氏完全佐劑與雙黃連注射液、生脈注射液聯(lián)用,對(duì)BN大鼠進(jìn)行致敏和激發(fā),比較聯(lián)用弗氏佐劑組與未加弗氏佐劑組動(dòng)物在過敏反應(yīng)發(fā)生率、過敏反應(yīng)發(fā)生程度;激發(fā)前后血漿中組胺、類胰蛋白酶、β-氨基己糖苷酶、Ig E含量及增長(zhǎng)率;被動(dòng)皮膚過敏試驗(yàn)大鼠皮下是否出現(xiàn)藍(lán)斑以及藍(lán)斑面積大小。結(jié)果:(1)聯(lián)用佐劑組過敏反應(yīng)的發(fā)生率以及過敏反應(yīng)的發(fā)生程度均高于未加佐劑組;(2)茵梔黃注射液組激發(fā)前后血漿中組胺、類胰蛋白酶、β-氨基己糖苷酶、Ig E的含量與生理鹽水組比較無顯著性差異;聯(lián)用佐劑后血漿中Ig E、組胺以及β-氨基己糖苷酶的含量均升高,與生理鹽水組相比具有顯著性的差異,激發(fā)前后四種指標(biāo)的變化率均高于茵梔黃注射液未加佐劑組;生脈注射液組激發(fā)前后血漿中四種指標(biāo)的含量與生理鹽水組比較無顯著性差異;聯(lián)用佐劑后血漿中Ig E、β-氨基己糖苷酶的含量均升高,與生理鹽水組相比具有顯著性的差異,激發(fā)前后四種指標(biāo)的變化率均高于生脈注射液未加佐劑組;(3)聯(lián)用佐劑組的被動(dòng)皮膚過敏反應(yīng)的陽性率均高于未加佐劑組。結(jié)論:弗氏完全佐劑可提高過敏反應(yīng)動(dòng)物模型的敏感性,有利于中藥注射劑安全性中過敏反應(yīng)的再評(píng)價(jià)。
[Abstract]:Objective: to observe the changes of sensitive indexes in the allergic reaction of Yinzhihuang injection, Shengmai injection adjuvant group and uncombined adjuvant group.To study the ability of this adjuvant to improve the stability and sensitivity of allergic reaction animal model in BN rats.Methods: BN rats were sensitized and stimulated by the combination of Freund's complete adjuvant and Shuanghuanglian injection and Shengmai injection. The incidence of allergic reaction and the degree of allergic reaction were compared between Freund's adjuvant group and non-Freund 's adjuvant group.Plasma histamine, trypsin, 尾 -aminohexosaminidase (尾 -glucosidase) IgE content and growth rate were detected before and after stimulation, and whether there were locus coeruleus and the size of locus coeruleus in passive skin allergy test rats.Results the incidence of allergic reaction and the degree of anaphylaxis in the adjuvant group were higher than those in the non-adjuvant group. The histamine in the plasma of the Yinzhihuang injection group was higher than that of the control group before and after stimulation.The change rate of the four indexes before and after stimulation was higher than that of Yinzhihuang injection without adjuvant group, and there was no significant difference between Shengmai injection group and physiological saline group before and after stimulation.The plasma levels of IgE, 尾-hexosaminidase increased after combined use of adjuvant, and there was a significant difference compared with the saline group.The change rates of the four indexes before and after stimulation were higher than those in the non-adjuvant group of Shengmai injection and the positive rate of the passive skin allergic reaction in the combined adjuvant group were higher than those in the non-adjuvant group.Conclusion: Freund's complete adjuvant can improve the sensitivity of allergic reaction animal model and be helpful to reevaluate the safety of traditional Chinese medicine injection.
