【摘要】：目的運(yùn)用生物信息學(xué)方法預(yù)測分析弓形蟲Toxofilin基因編碼蛋白的功能域及三級結(jié)構(gòu)。方法 PCR擴(kuò)增Toxofilin全長基因序列,利用SignaIP和TMHMM在線網(wǎng)站預(yù)測分析其編碼蛋白的信號肽、跨膜區(qū);利用DNAStar軟件分析Toxofilin蛋白的親水性、抗原指數(shù);聯(lián)合運(yùn)用ExPASY和PRODATA分析Toxofilin蛋白的功能域及三級結(jié)構(gòu)。結(jié)果 PCR擴(kuò)增Toxofilin基因大小為738bp。DNAstar分析其編碼蛋白Toxofilin序列存在典型信號肽,但不存在跨膜區(qū),在其N端存在較高的親水性基團(tuán)和抗原指數(shù)。ExPASY分析Toxofilin蛋白存在�；�、磷酸化蛋白激酶位點(diǎn)。結(jié)論 Toxofilin蛋白含有信號肽和磷酸化位點(diǎn)等功能域,抗原指數(shù),為驗(yàn)證Toxofilin功能奠定了理論基礎(chǔ)。
[Abstract]:Objective to predict and analyze the functional domain and tertiary structure of Toxofilin gene encoding protein of Toxoplasma gondii by bioinformatics. Methods the full-length Toxofilin gene sequence was amplified by PCR, and its signal peptide and transmembrane region were predicted and analyzed by SignaIP and TMHMM online website. DNAStar software was used to analyze the hydrophilicity and antigen index of Toxofilin protein, and ExPASY and PRODATA were used to analyze the functional domain and tertiary structure of Toxofilin protein. Results the size of Toxofilin gene amplified by PCR was a typical signal peptide in 738bp.DNAstar analysis of Toxofilin sequence of its encoded protein. However, there was no transmembrane region. There were high hydrophilic groups and antigen index. ExPASY analysis showed that Toxofilin protein had acylation and phosphorylated protein kinase site. Conclusion Toxofilin protein contains functional domains such as signal peptide and phosphorylation site, and antigen index. It lays a theoretical foundation for verifying the function of Toxofilin.
