血小板中自噬發(fā)生及功能研究

【摘要】:細(xì)胞自噬是細(xì)胞內(nèi)溶酶體依賴的降解和再循環(huán)過(guò)程,與許多生理和病理過(guò)程密切相關(guān)。雖然關(guān)于自噬調(diào)控機(jī)制的研究取得了很大進(jìn)展,但細(xì)胞核在自噬發(fā)生中的作用仍待研究。血小板在止血過(guò)程中發(fā)揮重要作用,是來(lái)源于巨核細(xì)胞的無(wú)核細(xì)胞,不存在基因轉(zhuǎn)錄。本研究利用血小板作為無(wú)核細(xì)胞模型研究自噬發(fā)生情況,討論細(xì)胞核在自噬發(fā)生中的作用。另一方面,本課題還研究了自噬對(duì)血小板功能的影響。我們發(fā)現(xiàn),血小板中存在自噬發(fā)生所需的相關(guān)機(jī)制;自噬泡可以被饑餓和Rapamycin誘導(dǎo)產(chǎn)生,且具有降解功能;血小板自噬的發(fā)生依賴Ⅲ型P13K的活性;自噬信號(hào)通路或自噬性降解受阻會(huì)下調(diào)血小板的聚集和粘附功能。結(jié)果提示,血小板中可以發(fā)生自噬,且自噬對(duì)于維持血小板的功能有重要作用;同時(shí)說(shuō)明,細(xì)胞核,至少?gòu)幕蜣D(zhuǎn)錄方面來(lái)說(shuō),對(duì)于自噬發(fā)生不是必須的。
[Abstract]:Autophagy is a lysosomal dependent degradation and recycling process in cells, which is closely related to many physiological and pathological processes. However, the role of nucleus in autophagy remains to be studied. Platelets play an important role in the hemostasis process. There is no gene transcription. In this study, we used platelets as a model to study autophagy and discuss the role of nucleus in autophagy. On the other hand, we also studied the effect of autophagy on platelet function.
