【摘要】：目的應(yīng)用改良的左冠狀動(dòng)脈回旋支結(jié)扎方法聯(lián)合微創(chuàng)手術(shù),制備小型豬急性心肌梗死模型,評(píng)估其有效性和穩(wěn)定性,并探討其在科研應(yīng)用中的意義。方法選擇3月齡巴馬香豬25頭,麻醉下行氣管插管呼吸機(jī)輔助通氣,微創(chuàng)開胸結(jié)扎左旋支中段,建立急性心肌梗死模型。并于術(shù)前、術(shù)后1 h、4周行心臟超聲評(píng)估模型動(dòng)物的心臟功能,術(shù)后4周處死動(dòng)物,取心臟行TTC染色評(píng)估梗死面積及病理變化,統(tǒng)計(jì)死亡率和死亡原因。結(jié)果結(jié)扎術(shù)后1 h和4周后心臟功能較術(shù)前均明顯降低,EF值由(64.2±4.6)%分別降至(48.2±5.3)%和(49.7±6.1)%(P0.01),左心室側(cè)壁室壁厚度變薄,膠原增生。梗死后1 h室顫率17.3%,梗死面積為(19.2±3)%。結(jié)論應(yīng)用改良的左冠狀動(dòng)脈回旋支結(jié)扎方法聯(lián)合微創(chuàng)手術(shù)能夠建立穩(wěn)定的豬心肌梗死模型,具有手術(shù)創(chuàng)傷小、模型穩(wěn)定和死亡率低等特點(diǎn),為相關(guān)研究提供了經(jīng)濟(jì)、理想的動(dòng)物模型。
[Abstract]:Objective to establish a mini-pig model of acute myocardial infarction by modified left coronary artery circumflex branch ligation combined with minimally invasive surgery, and to evaluate its efficacy and stability. Methods 25 Bama pigs aged 3 months were selected for tracheal intubation and ventilator assisted ventilation under anesthesia. The model of acute myocardial infarction was established by minimally invasive thoracotomy and ligation of left circumflex branch. The cardiac function of the model animals was evaluated by echocardiography at 1 hour and 4 weeks after operation, and the animals were killed 4 weeks after operation. The infarct size and pathological changes were evaluated by TTC staining. Results after 1 h and 4 weeks after ligation, the cardiac function decreased significantly from 64.2 鹵4.6% to 48.2 鹵5.3% and 49.7 鹵6.1%, respectively, and the left ventricular wall thickness became thinner. Collagen proliferation. The ventricular fibrillation rate was 17.3g / h after infarction, and the infarct area was 19.2 鹵30.Conclusion the modified left coronary circumflex branch ligation combined with minimally invasive surgery can establish a stable model of pig myocardial infarction with minimal surgical trauma. The characteristics of stable model and low mortality provide economic and ideal animal model for related research.
