家兔半膜肌各亞部肌球蛋白重鏈表達(dá)差異的實(shí)驗(yàn)研究

【摘要】:目的骨骼肌肌纖維類型的構(gòu)成比較復(fù)雜,呈單一性分布的較少。人們發(fā)現(xiàn)家兔半膜肌各個(gè)亞部的肌纖維構(gòu)成較其它骨骼肌單一。本實(shí)驗(yàn)在此基礎(chǔ)上通過肌球蛋白重鏈的亞型的分類方法檢測家兔半膜肌各亞部的肌纖維型的構(gòu)成情況,從分子水平觀察其肌纖維類型的分布特點(diǎn),為進(jìn)一步研究兩型肌纖維在結(jié)構(gòu)、能量代謝、基因表達(dá)和功能等方面的區(qū)別以及兩型肌纖維轉(zhuǎn)化機(jī)制奠定基礎(chǔ)。 方法實(shí)驗(yàn)以10只成年家兔為研究對象,雌雄不拘,取其右側(cè)肢體半膜肌。通過改良肌球蛋白ATP酶染色的方法驗(yàn)證其四個(gè)亞部兩型肌纖維分布情況及構(gòu)成比例,同時(shí)采用反轉(zhuǎn)錄聚合酶鏈?zhǔn)椒磻?yīng)(RT-PCR)檢測各個(gè)亞部MHCⅠ、MHCⅡa、MHCⅡb、及MHCⅡx四個(gè)亞型mRNA的構(gòu)成及含量。 結(jié)果家兔半膜肌四個(gè)亞部中,固有半膜肌只含有慢肌(Ⅰ型肌纖維),表達(dá)MHCⅠ型和少量MHCⅡa型基因。副半膜肌腹側(cè)含有少量慢肌(Ⅰ型肌纖維),表達(dá)少量MHCⅠ型基因,其余主要含有快肌(Ⅱ型肌纖維),表達(dá)MHCⅡa、MHCⅡb和MHCⅡx型基因;而其背側(cè)和外側(cè)只含有快肌,僅表達(dá)MHCⅡa、MHCⅡb和MHCⅡx型基因。 結(jié)論固有半膜肌亞部為慢肌,含有慢肌纖維,即Ⅰ型肌纖維,主要表達(dá)MHCⅠ和MHCⅡa兩個(gè)亞型。副半膜肌的三個(gè)亞部中,除腹側(cè)亞部表達(dá)極少量MHCⅠ亞型,均主要表達(dá)MHCⅡa、MHCⅡb和MHCⅡx亞型。背側(cè)和外側(cè)亞部不含有慢肌纖維(Ⅰ型肌纖維),不表達(dá)MHCⅠ亞型。
[Abstract]:Objective the types of skeletal muscle fibers are complicated. It was found that the muscle fiber composition of rabbit semilemma muscle was simpler than that of other skeletal muscles. On the basis of this, the myosin heavy chain subtype classification method was used to detect the semilemma muscle of rabbits. The composition of the myofibrillar type of the subpart, The distribution characteristics of muscle fiber types were observed at the molecular level, which laid a foundation for further study on the differences in structure, energy metabolism, gene expression and function between the two types of muscle fibers and the mechanism of transformation of the two types of muscle fibers.
