三味干姜散對兩種實驗性肝損動物模型的調(diào)控作用研究

【摘要】:目的從保護肝臟線粒體和調(diào)控"Nrf2/Bach1-ARE-抗氧化酶"通路兩個角度,觀察三味干姜散對實驗性肝損傷動物模型的影響。方法復制大鼠急性酒精性肝損傷模型和小鼠刀豆蛋白A(Con A)免疫性肝損傷模型,分別給予三味干姜散后,檢測肝功能指標ALT、AST,進行病理學觀察;提取肝線粒體,檢測線粒體膜電位、MPTP、SOD、8-OHd G、呼吸鏈復合物Ⅰ、Ⅲ酶活性以及肝組織中MDA、GSH、GSH-Px水平;采用RT-PCR檢測肝組織中Nrf2、Bach1、HO-1的m RNA表達水平。結(jié)果對于急性酒精性肝損傷模型,三味干姜散能降低ALT、AST(P0.05)和8-OHd G(P0.01)以及MDA的含量(P0.05,P0.01),提高肝組織中抗氧化劑GSH、SOD和GSH-Px酶的活性(P0.05,P0.01),上調(diào)Nrf2和HO-1 m RNA的表達(P0.05),顯著增加呼吸鏈復合物Ⅰ、Ⅲ酶的活性(P0.01),以及降低線粒體膜通道孔的通透性(P0.05);對于免疫性肝損傷模型,三味干姜散能降低ALT、AST水平和MDA活性(P0.05,P0.01),升高SOD(P0.05)、GSH-Px與GSH酶的活性(P0.05,P0.01),顯著上調(diào)線粒體膜電位(P0.01)。結(jié)論三味干姜散對大鼠急性酒精性肝損傷和小鼠Con A免疫性肝損傷均有一定的保護作用,其機制與三味干姜散抗肝臟線粒體損傷和調(diào)控"Nrf2/Bach1-ARE-抗氧化酶"通路有關(guān)。
[Abstract]:Objective to observe the effect of Sanwei Ganjiang Powder on experimental liver injury from two aspects: protecting liver mitochondria and regulating "Nrf2 / Bach1-ARE- antioxidant enzyme" pathway.Methods the acute alcoholic liver injury model in rats and the immunological liver injury model in mice with concanavalin A(Con A were established. The liver function indexes of alt AST were detected and the liver mitochondria were extracted after three kinds of dry ginger powder were given respectively.Conclusion Sanwei Ganjiang Powder has protective effects on acute alcoholic liver injury in rats and Con A immune liver injury in mice, and its mechanism is related to the regulation of "Nrf2 / Bach1-ARE- antioxidant enzyme" pathway by Sanwei Ganjiang Powder in preventing liver mitochondria damage and in regulating "Nrf2 / Bach1-ARE- antioxidant enzyme" pathway.
