【摘要】：目的:研究消退素D1(Resolvin D1,Rv D1)對(duì)非壓迫性腰椎間盤(pán)突出癥模型大鼠背根神經(jīng)節(jié)(dorsal root ganglion,DRG)炎癥的影響及內(nèi)在機(jī)制。方法:選取30只雄性大鼠建立非壓迫性腰椎間盤(pán)突出癥模型。隨機(jī)分為假手術(shù)組、模型組、Rv D1組(100 ng)。術(shù)后連續(xù)3天模型組和Rv D1組分別鞘內(nèi)注射10μl磷酸鹽緩沖液、Rv D1。于術(shù)前1天及術(shù)后連續(xù)7天檢測(cè)各組大鼠術(shù)側(cè)50%縮足閾值(50%paw withdrawal threshold,PWT)。術(shù)后第7天,酶聯(lián)免疫吸附測(cè)定術(shù)側(cè)L5 DRG中TNF-α和IL-1β的蛋白表達(dá)量,免疫組化法檢測(cè)DRG中p-ERK及NF-κB/p65的陽(yáng)性表達(dá)。結(jié)果:術(shù)后各時(shí)間點(diǎn),模型組大鼠50%PWT較假手術(shù)組明顯降低(P0.01);TNF-α和IL-1β表達(dá)明顯升高(P0.01);p-ERK、NF-κB/p65蛋白水平明顯升高(P0.01)。術(shù)后第2~7天,Rv D1組與模型組相比,50%PWT顯著升高(P0.05);TNF-α和IL-1β的蛋白表達(dá)明顯降低(P0.01);p-ERK及NF-κB/p65蛋白水平明顯降低(P0.01)。結(jié)論:Rv D1促進(jìn)背根神經(jīng)節(jié)炎癥的消退,可能與調(diào)節(jié)炎癥介質(zhì)TNF-α和IL-1β的表達(dá)及ERK和NF-κB/p65通路活性有關(guān)。
[Abstract]:Objective : To study the effects and internal mechanisms of melatonin D1 ( Rv D1 ) on the inflammation of dorsal root ganglion ( DRG ) in rats with non - compression lumbar disc herniation . Methods : Thirty male rats were selected to establish a model of non - compression lumbar disc herniation . Ten 渭l of phosphate buffer solution and Rv D1 were injected into the sheath of the 3 - day and Rv - D1 groups after operation , and the 50 % paw withdrawal threshold was detected in each group on the first day before operation and 7 days after operation . Results : The expression of TNF - 偽 and IL - 1尾 in DRG was significantly decreased ( P0.01 ) . The expression of TNF - 偽 and IL - 1尾 in DRG increased significantly ( P0.01 ) , and the expression of TNF - 偽 and IL - 1尾 in DRG was significantly decreased ( P0.01 ) .
