紫草油膏對(duì)褥瘡模型大鼠的治療作用及機(jī)制

【摘要】:目的:觀察紫草油膏對(duì)褥瘡模型大鼠的治療作用及可能的作用機(jī)制,為其臨床推廣應(yīng)用提供一定的實(shí)驗(yàn)依據(jù)。方法:采用皮下植入鐵片加磁鐵的方法復(fù)制大鼠褥瘡模型。將造模成功的大鼠隨機(jī)分為模型組、陽性(消炎生肌膏)組、紫草油膏中、高劑量組和碘劑組。另設(shè)空白組和假手術(shù)組。于造模成功后次日開始外涂給藥,每日1次。連續(xù)給藥3 d后,肉眼觀察受壓部位皮膚形態(tài)學(xué)和顏色變化;光鏡下觀察受壓部位皮膚組織病理學(xué)變化;免疫組化法檢測核轉(zhuǎn)錄因子-κB p65(nuclear transcription factor-κB,NF-κB p65)表達(dá)水平;水溶性四唑鹽法及微板法檢測大鼠血清中超氧化物歧化酶(superoxide dismutase,SOD),丙二醛(malondialdehyde,MDA)的含量;酶聯(lián)免疫吸附法(enzyme linked immunosorbent assay,ELISA)測定大鼠血清中白細(xì)胞介素-lα(interleukin-lα,IL-1α),腫瘤壞死因子-α(tumour necrosis factorα-α,TNF-α)的含量。結(jié)果:與空白組及假手術(shù)組比較,模型組大鼠受壓部位發(fā)紅不改變;病理觀察到細(xì)胞排列紊亂,細(xì)胞間隙增寬,炎癥細(xì)胞浸潤等現(xiàn)象;血清中MDA,TNF-α,IL-1α及皮膚組織細(xì)胞核中NF-κB p65的含量明顯升高(P0.05,P0.01),而血清中SOD的含量顯著降低(P0.01)。紫草油膏治療3 d后,與模型組比較,受壓部位皮膚組織紅腫消退,狀態(tài)明顯好轉(zhuǎn);病理狀態(tài)明顯得到改善,炎癥程度和數(shù)量均明顯減輕或減少。紫草油膏中、高劑量組大鼠血清中SOD的含量明顯高于模型組,而MDA的含量明顯低于模型組(P0.05,P0.01);紫草油膏高劑量組較模型組血清中TNF-α,IL-1α的含量顯著降低(P0.05),細(xì)胞核中NF-κB p65蛋白表達(dá)顯著降低(P0.05)。結(jié)論:紫草油膏對(duì)褥瘡模型大鼠具有一定治療作用,其機(jī)制與抑制細(xì)胞核中NF-κB p65的表達(dá)、降低大鼠血清中IL-lα,TNF-α,MDA的含量及增加SOD含量有關(guān)。
[Abstract]:Objective: to observe the therapeutic effect and possible mechanism of purple herb ointment on bedsore model rats. Methods: the rat model of bedsore was established by subcutaneous implantation of iron and magnet. The successful rats were randomly divided into three groups: model group, positive group (Xiaoyan Shengji ointment), and purple herb ointment. The high dose group and iodine group were divided into blank group and sham operation group. The next day after the model was successfully made, the drug was applied externally, once a day. After 3 days of continuous administration, the changes of skin morphology and color were observed with the naked eye. The histopathological changes of skin were observed under light microscope, the expression of nuclear transcription factor-魏 B p65(nuclear transcription factor- 魏 B (NF- 魏 B p65) was detected by immunohistochemical method, and the content of superoxide dismutase (SOD) and malondialdehyde (MDA) in serum of rats were detected by water-soluble tetrazolium and microplate method. The contents of interleukin-l 偽 -interleukin-l 偽 -guanil IL-1 偽, tumor necrosis factor- 偽 tumour necrosis factor 偽-偽 -TNF- 偽 in serum of rats were determined by enzyme linked immunosorbent assay-ELISA.Results: compared with the control group and sham operation group, the rats in the model group did not change the redness of the pressed parts. Pathological observation showed that the cell arrangement was disordered, the gap widened and inflammatory cell infiltrated, the contents of MDA-TNF- 偽 IL-1 偽 in serum and NF- 魏 B p65 in skin tissue nucleus increased significantly, while the content of SOD in serum decreased significantly after 3 days of treatment. Compared with the model group, the redness and swelling of the skin tissue in the compressed area receded, the pathological state was obviously improved, the degree of inflammation and the amount of inflammation were significantly reduced or decreased. The content of serum SOD in high dose group was significantly higher than that in model group. The content of MDA was significantly lower than that of the model group, the content of TNF- 偽 and IL-1 偽 in serum of the model group was significantly lower than that of the model group, and the expression of NF- 魏 B p65 protein in the nucleus of the model group was significantly lower than that of the model group. Conclusion: YC ointment has a certain therapeutic effect on the model rats with bedsore. The mechanism is related to the inhibition of the expression of NF- 魏 B p65 in the nucleus, the decrease of the content of IL-l 偽 -TNF- 偽 and the increase of the content of SOD in the serum of rats.
