外源性一氧化氮對(duì)旋毛蟲感染小鼠抗氧化能力的影響

【摘要】:目的探討外源性NO供體亞硝基鐵氰化鈉(SNP)對(duì)旋毛蟲病小鼠肝臟、外周血中抗氧化酶活性及脂質(zhì)氧化的影響。方法采用消化法分離旋毛蟲肌幼蟲,感染BALB/c小鼠,400條/只。42 d后,收集感染鼠及正常小鼠外周血、肝臟。設(shè)A組(感染鼠肝臟勻漿物+SNP)、B組(正常鼠肝臟勻漿物+SNP)、C組(感染鼠外周血+SNP)、D組(正常鼠外周血+SNP)。每組內(nèi)設(shè)5個(gè)SNP反應(yīng)終濃度,分別為0(空白對(duì)照)、2、5、10、30μmol/L,37℃反應(yīng)30 min,測(cè)定各組超氧化物歧化酶(SOD)、過(guò)氧化氫酶(CAT)、谷胱甘肽過(guò)氧化物酶(GSH-Px)及脂質(zhì)氧化(MDA)含量。結(jié)果與正常鼠相比,旋毛蟲感染鼠肝臟、外周血中SOD、CAT、GSH-Px活性上升,MDA含量增加,差異均有統(tǒng)計(jì)學(xué)意義(P均0.05);在A、B組中,10、30μmol/L SNP作用下SOD、CAT、GSH-Px活性較空白對(duì)照降低(P均0.05);與2~30μmol/L SNP反應(yīng)后,MDA含量較空白對(duì)照明顯升高(P均0.05)。在C、D組中,30μmol/LSNP作用下SOD、CAT、GSH-Px活性顯著下降,MDA濃度升高,與空白對(duì)照比較差異均有統(tǒng)計(jì)學(xué)意義(P均0.01)。在2~30μmol/L濃度區(qū)間,SNP濃度與旋毛蟲感染鼠肝臟、外周血中SOD、CAT、GSH-Px活性呈負(fù)相關(guān)(r肝SOD=-0.901,r肝CAT=-0.802,r肝GSH-Px=-0.847,r血SOD=-0.899,r血CAT=-0.685,r血GSH-Px=-0.635,P均0.05),與MDA含量呈正相關(guān)(r肝MDA=0.697,r血MDA=0.764,P均0.05)。結(jié)論旋毛蟲感染可致小鼠肝臟、外周血中活性氧自由基產(chǎn)生增加,SOD、CAT、GSH-Px活性升高;外源性NO可降低旋毛蟲感染小鼠SOD、CAT、GSH-Px活性,抑制小鼠的抗氧化能力。
[Abstract]:Objective to investigate the effects of exogenous no donor sodium nitroso cyanide (SNPs) on the activities of antioxidant enzymes and lipid oxidation in the liver and peripheral blood of mice with trichinellosis. Methods the muscle larvae of Trichinella spiralis were isolated by digestion and infected with 400 caterpillar larvae per mouse for 42 days.
