基于氣機升降理論觀察旋覆代赭湯及其拆方對反流性食管炎模型大鼠食管線粒體ANT轉(zhuǎn)運活性的影響

【摘要】:目的:觀察旋覆代赭湯及其拆方對反流性食管炎(RE)模型大鼠線粒體腺苷酸轉(zhuǎn)運體(ANT)轉(zhuǎn)運活性的影響。方法:將60只雄性Wistar大鼠,隨機分成6組,即正常組、模型組、西藥組,旋覆代赭湯全方組及拆方組(包括苦降組、甘升組),每組10只大鼠。對除正常組外的造模組大鼠采用"4.2 mm幽門夾+胃底2/3結(jié)扎術(shù)"制備RE動物模型。從術(shù)后第7天開始,正常對照組、模型對照組給予生理鹽水灌胃,旋覆代赭湯全方組、拆方各組及西藥組分別給予相應(yīng)藥液灌胃,干預(yù)14 d后,第22天處死全部大鼠后,取材,在光學(xué)顯微鏡下觀察RE大鼠病理分級,各組隨機取6份食管標本勻漿制成線粒體懸浮,液通過H3-ADP摻入法測量檢測食管線粒體ANT轉(zhuǎn)運活性。結(jié)果:各組大鼠食管黏膜病理形態(tài)學(xué)積分比較:模型組大鼠食管黏膜病理積分比正常組顯著升高,有顯著性差異(P0.01);全方組、西藥組、甘升組食管黏膜病理形態(tài)學(xué)積分比模型組明顯降低,差異有統(tǒng)計學(xué)意義(P0.05),苦降組食管黏膜病理形態(tài)學(xué)積分較模型組稍有降低,但差異無統(tǒng)計學(xué)意義(P0.05);全方組及西藥組食管黏膜病理形態(tài)學(xué)積分較甘升組稍降低,但差異無統(tǒng)計學(xué)意義(P0.05)。各組食管線粒體ANT轉(zhuǎn)運活性比較:與正常組比較,模型組、西藥組、全方組、甘升組大鼠食管線粒體ANT轉(zhuǎn)運活性略提高,差異無統(tǒng)計學(xué)意義(P0.05),苦降組略降低,差異無統(tǒng)計學(xué)意義(P0.05);與模型組比較,西藥組、全方組及甘升組大鼠食管線粒體ANT轉(zhuǎn)運活性略提高略升高,但差異無統(tǒng)計學(xué)意義(P0.05);西藥組、全方組、甘升組及苦降組組間比較均無統(tǒng)計學(xué)意義(P0.05)。結(jié)論:旋覆代赭湯通過調(diào)節(jié)脾胃氣機改善RE模型大鼠食管黏膜光鏡下表現(xiàn),促進黏膜修復(fù),改善炎癥。在一定程度上提高RE大鼠線粒體ANT轉(zhuǎn)運活性,輔助線粒體能量的順利完成,亦可能是旋覆代赭湯治療RE的作用機制之一。
[Abstract]:Objective: to observe the effect of Xuanhudaizhe decoction and its disassembly on mitochondrial adenylate transporter (ANT) transport activity in rat model of reflux esophagitis. Methods: sixty male Wistar rats were randomly divided into 6 groups: normal group, model group and western medicine group. Xuanhudezhe decoction group and disassembly group (including Kujiang group, Gansheng group, 10 rats in each group). The rats in the model group other than the normal group were treated with "4.2 mm pyloric clamp stomach fundus 2 / 3 ligation" to make RE animal model. From the 7th day after operation, RE animal model was established. The normal control group and the model control group were given normal saline intragastric perfusion, Xinfen daizhe decoction group, the disassembled prescription group and western medicine group respectively. After 14 days of intervention, all the rats were killed on the 22nd day, and the materials were obtained. The pathological grades of RE rats were observed under optical microscope. Six esophageal homogenates were randomly selected for mitochondrial suspension in each group. Results: compared with the normal group, the pathological score of esophageal mucosa in the model group was significantly higher than that in the normal group (P 0.01). In the western medicine group, the pathological morphology integral of the esophageal mucosa in the Gan-sheng group was significantly lower than that in the model group, and the difference was statistically significant (P 0.05). The pathological morphology integral of the esophageal mucosa in the Kujiang group was slightly lower than that in the model group. But the difference was not statistically significant (P 0.05), the score of pathomorphology of esophageal mucosa in the whole prescription group and the western medicine group was slightly lower than that in the Gan-sheng group, but the difference was not statistically significant (P 0.05). The comparison of the ANT transport activity of the esophageal mitochondria in each group: compared with the normal group, the model group, the western medicine group, the control group, the model group and the western medicine group, In the whole prescription group, the mitochondrial ANT transport activity of the rats in the Gan-sheng group increased slightly, but the difference was not statistically significant (P 0.05), but the difference was not significant in the bitter group (P 0.05), and compared with the model group, the western medicine group, the western medicine group, The activity of mitochondrial ANT transport increased slightly in the whole prescription group and the Gansheng group, but the difference was not statistically significant (P 0.05), while that in the western medicine group, the whole prescription group, the western medicine group, the whole prescription group, the western medicine group, the whole prescription group, There was no significant difference between Gan-sheng group and Ku-Jiang group (P 0.05). Conclusion: Xuanfudezhe decoction can improve the esophageal mucosal light microscope performance and promote mucosal repair by regulating spleen and stomach Qi. To improve inflammation, to some extent increase the activity of mitochondrial ANT transport in RE rats and assist the smooth completion of mitochondrial energy, which may also be one of the mechanisms of Xuanhudaizhe decoction in the treatment of RE.
