【摘要】:基因轉(zhuǎn)錄是一個(gè)有序的多步驟過程,包括轉(zhuǎn)錄起始前、轉(zhuǎn)錄起始和轉(zhuǎn)錄延伸等幾個(gè)階段。長(zhǎng)期以來,轉(zhuǎn)錄起始前及轉(zhuǎn)錄起始階段調(diào)控一直被認(rèn)為是基因轉(zhuǎn)錄的關(guān)鍵限速步驟。然而,近年來利用全基因組深度測(cè)序等新技術(shù)對(duì)轉(zhuǎn)錄過程的深入研究,發(fā)現(xiàn)轉(zhuǎn)錄起始后的轉(zhuǎn)錄延伸階段也是許多基因表達(dá)調(diào)控的關(guān)鍵步驟,并且這種調(diào)控機(jī)制在從果蠅到哺乳動(dòng)物的不同物種之間高度保守。本文綜述了NELF、DSIF和P-TEFb等主要轉(zhuǎn)錄延伸調(diào)控因子調(diào)控RNA聚合酶Ⅱ轉(zhuǎn)錄暫停和高效轉(zhuǎn)錄延伸最新分子機(jī)制,并討論了轉(zhuǎn)錄延伸調(diào)控在癌癥、炎癥以及病毒感染等疾病發(fā)生中的作用。
[Abstract]:Gene transcription is an orderly multistep process that includes several stages, including pre-transcriptional initiation, transcriptional initiation and transcriptional extension. The regulation of pretranscriptional and transcriptional initiation stages has been considered to be a key rate-limiting step in gene transcription. However, in recent years, novel techniques such as full-genome deep sequencing have been used to further study the transcription process. It has been found that the transcriptional extension after transcription initiation is also a key step in the regulation of many genes expression.
