兩種鼓室硬化造模方法的比較

【摘要】:目的應(yīng)用鼓膜切開法和鼓室接種細(xì)菌法構(gòu)建鼓室硬化動(dòng)物模型,觀察兩種方法造模后其鼓膜和鼓室粘膜的組織形態(tài)學(xué)變化。方法 40只SD大鼠分為4組,每組10只:A1組雙耳鼓膜不作處理,作為正常對(duì)照;A2組雙耳鼓膜緊張部后下象限作2mm的切口;B1組雙側(cè)鼓室不作處理,作為正常對(duì)照;B2組雙側(cè)鼓室接種1×108 CFU/ml肺炎鏈球菌0.1ml。分別于鼓膜切開后第2周及鼓室接種肺炎鏈球菌后第1、2、4、6、8周時(shí)觀察各組大鼠鼓膜情況,比較鼓室硬化發(fā)生率;分別于造模后第2周和第8周取材行聽(tīng)泡連續(xù)切片HE染色,觀察鼓膜及鼓室粘膜的組織形態(tài)學(xué)變化。結(jié)果 A2組14耳發(fā)生鼓膜鈣化(14/20,70%);B2組死亡一只,6耳發(fā)生鼓膜鈣化(6/18,33.33%),兩組的鼓膜及鼓室粘膜均可觀察到炎癥細(xì)胞浸潤(rùn)及纖維組織增生,但B2組的改變更顯著。兩對(duì)照組(A1和B1組)既未發(fā)生鼓膜鈣化,也未出現(xiàn)組織形態(tài)學(xué)改變。結(jié)論兩種鼓室硬化造模方法均切實(shí)可行,但鼓膜切開法操作更簡(jiǎn)單,成模周期更短,成模率更高。
[Abstract]:Objective application of myringotomy method and tympanic inoculation method to construct animal model of tympanosclerosis, observation of two methods after modeling the tympanic membrane and tympanic mucosa tissue morphologic changes. Methods 40 SD rats were divided into 4 groups, 10 rats in each group: A1 group mtau remaineduntreated as normal control; group A2. Tensa posterior inferior quadrant 2mm incision; group B1 bilateral tympanic remaineduntreated as normal control; group B2 bilateral tympanic inoculation of 1 * 108 CFU/ml of Streptococcus pneumoniae 0.1ml. respectively in myringotomy in 1,2,4,6,8 weeks after second weeks after inoculation of Streptococcus pneumoniae and tympanic cavity were observed in rat tympanic membrane, the incidence of tympanosclerosis in line; second weeks and eighth weeks after listening to the global serial sections of HE staining, the morphological changes of the tympanic membrane and the tympanic mucosa was observed. Results in A2 group, 14 ears of tympanic membrane calcification (14/20,70%) occurred in B2 group; death one, 6 ears occurred The tympanic membrane calcification (6/18,33.33%), and the tympanic membrane tympanic mucosa can be observed two groups of inflammatory cell infiltration and hyperplasia of fibrous tissue, but the B2 group changed more significantly. Two control groups (A1 and B1 group) did not happen tympanic membrane calcification, did not appear morphological changes. Conclusion two kinds of tympanosclerosis modeling methods are feasible but, myringotomy method more simple operation, shorter cycle model, the model rate higher.
