【摘要】：目的研究枸杞多糖(LBP)對(duì)生精功能和生精干細(xì)胞的影響。方法 122只雄性昆明小鼠中隨機(jī)選取92只,通過(guò)35 mg/kg白消安造模,獲得生精障礙動(dòng)物模型,另外30只小鼠為正常對(duì)照組。40 d后對(duì)造模成功的小鼠隨機(jī)分為自然恢復(fù)組(n=23)和LBP低劑量組(n=16,75 mg/kg)、中劑量組(n=19,150 mg/kg)、高劑量組(n=16,300 mg/kg),連續(xù)灌胃給藥70 d(小鼠2個(gè)生精周期)后進(jìn)行觀察,并與正常對(duì)照組進(jìn)行比較,觀察其生理狀態(tài)和生殖功能變化。結(jié)果 LBP在造模動(dòng)物的外觀體征、機(jī)能和行為的表現(xiàn)、動(dòng)物正常發(fā)育狀態(tài)(體質(zhì)量)、生殖器官組織重量(睪丸、附睪和儲(chǔ)精囊、前列腺-會(huì)陰復(fù)合體)、附睪精子質(zhì)量、睪丸組織精子生成量、睪丸組織形態(tài)學(xué)和c-kit表達(dá)量、精曲小管直徑和空泡化程度、生精上皮厚度和細(xì)胞層數(shù)、c-kit陽(yáng)性細(xì)胞數(shù)、H-score評(píng)分、平均吸光度(IA)等系列指標(biāo)均顯示明顯高于自然恢復(fù)組(P0.05,P0.01),并具有劑量效應(yīng)關(guān)系,但仍未達(dá)到正常對(duì)照動(dòng)物的水平。結(jié)論 LBP具有明顯促進(jìn)白消安生精障礙模型小鼠的生精能力的作用,可以顯著增加生精干細(xì)胞、附睪精子質(zhì)量和睪丸精子生成量。
[Abstract]:Objective to study the effect of Lycium barbarum polysaccharides (LBP) on spermatogenic function and spermatogenic stem cells. The other 30 mice were randomly divided into natural recovery group (n = 23) and LBP low dose group (n = 1675 mg / kg 路kg ~ (-1)), middle dose group (n = 19150 mg / kg) and high dose group (n = 16) (n = 16), 300 mg 路kg ~ (-1) 路kg ~ (-1) 路kg ~ (-1) for 70 d (2 spermatogenic cycles). Results the appearance, function and behavior of LBP in model animals, normal developmental state (body weight, weight of reproductive organs, testis, testis) were observed and compared with those in normal control group. Epididymis and seminal vesicle, prostate-perineum complex, epididymal sperm quality, testicular sperm production, testicular morphology and c-kit expression, seminiferous tubule diameter and degree of vacuolation. The thickness of spermatogenic epithelium and the number of c-kit positive cells in spermatogenic epithelium and the number of c-kit positive cells were significantly higher than those in natural recovery group. Conclusion LBP can promote the spermatogenic ability of mice with spermatogenic disorder, and can significantly increase the spermatogenic stem cells, the sperm quality of epididymis and the quantity of testicular spermatozoa.
