束縛-應(yīng)激聯(lián)合辣素灌胃建立大鼠腹瀉型腸易激綜合征模型及評(píng)價(jià)

【摘要】:目的基于觀察大鼠癥狀學(xué)、組織病理學(xué)、內(nèi)臟敏感性、肥大細(xì)胞活化、自噬、Beclin-1及Claudin-2等相關(guān)指標(biāo)變化,評(píng)價(jià)束縛-應(yīng)激聯(lián)合辣素灌胃建立一種新的腹瀉型腸易激綜合征(diarrhea-predominant irritable bowel syndrome,D-IBS)模型研究。方法清潔級(jí)雄性SD大鼠40只,按體重分為正常組、模型I組、模型II組及模型III組,每組10只。模型組均采用束縛-夾尾應(yīng)激聯(lián)合辣素(I組0.125%、II組0.250%、III組0.500%)灌胃制備D-IBS模型,正常組予等劑量生理鹽水灌胃處理,連續(xù)2周。分別應(yīng)用Power lab儀檢測(cè)大鼠腹壁收縮次數(shù)及弓背次數(shù)變化,肥大細(xì)胞染色液(醛品紅-橙黃G法)檢測(cè)肥大細(xì)胞活化情況,采用光鏡、電鏡檢測(cè)大鼠結(jié)腸組織病理學(xué)及其自噬變化,應(yīng)用免疫組化SABC法檢測(cè)大鼠結(jié)腸黏膜Beclin-1及Claudin-2表達(dá)變化。結(jié)果模型III組大鼠全部死亡,模型II組大鼠大便次數(shù)增多,內(nèi)臟敏感性閾值降低,與正常組及模型I組比較,差異有顯著性(P0.05);各組大鼠結(jié)腸黏膜、黏膜上皮絨毛及腺體形態(tài)正常,未見(jiàn)黏膜下血管擴(kuò)張及彌漫性炎癥浸潤(rùn)改變;正常組除外,模型I組、II組大鼠結(jié)腸組織黏膜間質(zhì)或黏膜下層可見(jiàn)類圓形紫紅至深紫色染色點(diǎn),提示肥大細(xì)胞表達(dá)增多;模型II組大鼠結(jié)腸腸上皮細(xì)胞自噬、Beclin-1及Claudin-2表達(dá)明顯增多,與正常組及模型I組相比,差異有顯著性(P0.05)。結(jié)論束縛-應(yīng)激聯(lián)合0.25%辣素灌胃建立的大鼠D-IBS模型以腹瀉增多、內(nèi)臟高敏感、肥大細(xì)胞表達(dá)增高、腸上皮細(xì)胞自噬增加、腸黏膜屏障破壞等表現(xiàn)為特點(diǎn),與人IBS較為相似,該方法簡(jiǎn)單易行,值得推廣應(yīng)用。
[Abstract]:Objective to observe the changes of symptoms, histopathology, visceral sensitivity, mast cell activation, autophagy Beclin-1 and Claudin-2 in rats. To evaluate the effect of restraint stress combined with pepsin on the establishment of a new model of diarrhea irritable irritable bowel syndrome D-IBSs. Methods Forty clean male SD rats were divided into normal group, model group I, model group II and model III group according to their body weight. The D-IBS model was made by intragastric administration of restraint and tail stress combined with capsaicin I group (0.125%) and normal saline group (0.500%) in group II (group II) and normal saline group (n = 10), and normal control group (n = 10), and normal group (n = 10) were treated by intragastric administration of normal saline at the same dose. For 2 weeks, the changes of the number of contraction of abdominal wall and the number of times of arched back were detected by Power lab instrument, and the activation of mast cells in mast cell staining solution (aldehyde fuchsin orange G method) was detected by light microscope. The changes of colonic histopathology and autophagy were examined by electron microscope, and the expression of Beclin-1 and Claudin-2 in colonic mucosa of rats were detected by immunohistochemical SABC method. Results all the rats in model III group died and the defecation times in model group II increased. The visceral sensitivity threshold was significantly lower than that in normal group and model group I, the colonic mucosa, epithelial villi and glands were normal in each group, no submucosal vasodilation and diffuse inflammatory infiltration were observed. Except the normal group, the colonic mucosa stroma or submucosa of the model group I and II showed round purplish red to dark purple staining points, indicating the increase of mast cell expression. Compared with normal group and model I group, the expression of Beclin-1 and Claudin-2 in colonic epithelial cells of model II rats was significantly higher than that in normal group and model I group. Conclusion the D-IBS model of rats induced by restraint stress combined with 0.25% caprine is more diarrhea. The hypersensitivity of viscera, the increase of mast cell expression, the increase of intestinal epithelial cell autophagy and the destruction of intestinal mucosal barrier are characteristic features, which are similar to human IBS. This method is simple and easy to use and is worth popularizing.
