電針委中穴對腰椎退變模型大鼠纖維環(huán)細(xì)胞caspase3、XIAP蛋白表達(dá)的影響

【摘要】:目的:觀察電針委中穴對腰椎退變模型大鼠纖維環(huán)細(xì)胞凋亡及caspase3和XIAP表達(dá)的影響。方法:將40只雄性SD大鼠按隨機(jī)數(shù)字表法分為假手術(shù)組、模型組、電針委中穴組、電針非穴組。采用纖維環(huán)穿刺法制作腰椎退變模型。于造模1個月后行電針治療,電針委中穴組取"委中"穴,電針非穴組取委中內(nèi)側(cè)0.5cm,均每日1次,每次20min,連續(xù)治療4周。治療結(jié)束后取材,運(yùn)用TUNEL法檢測纖維環(huán)細(xì)胞凋亡,免疫組化檢測(caspase3)和XIAP表達(dá)。結(jié)果:假手術(shù)組偶見TUNEL細(xì)胞,caspase3低表達(dá),XIAP高表達(dá);模型組纖維環(huán)細(xì)胞TUENL陽性細(xì)胞和caspase3表達(dá)較假手術(shù)組顯著增加(P0.01),XIAP表達(dá)低于假手術(shù)組(P0.01);與模型組比較,電針委中穴組TUNEL陽性細(xì)胞和caspase3表達(dá)均明顯減少(P0.01),XIAP表達(dá)增加(P0.01);與電針委中穴組比較,電針非穴組纖維環(huán)細(xì)胞TUENL陽性細(xì)胞和caspase3表達(dá)較高(P0.05),XIAP蛋白表達(dá)減少(P0.05)。結(jié)論:電針可延緩椎間盤退變,其機(jī)制可能與調(diào)節(jié)纖維環(huán)細(xì)胞caspase3和XIAP表達(dá),抑制纖維環(huán)細(xì)胞凋亡有關(guān)。
[Abstract]:Objective: to observe the effect of electroacupuncture (EA) on apoptosis and expression of caspase3 and XIAP in fibrocytic cells of lumbar degenerative rats. Methods: forty male Sprague-Dawley rats were randomly divided into sham-operation group, model group and EA Weizhong point group. The model of lumbar degenerative degeneration was made by using fiber loop puncture method. Electroacupuncture treatment was performed 1 month later. The acupoints of "Wei Zhong" in the acupoint group and 0.5 cm on the medial side in the non-point group were obtained once a day. After 20 min treatment for 4 weeks, the apoptosis of fibrous ring cells was detected by TUNEL method, and the expression of caspase3) and XIAP were detected by immunohistochemistry. Results: in sham-operation group, the low expression of TUNEL cell caspase3 and the high expression of XIAP were occasionally observed. The expression of TUENL positive cells and caspase3 in the model group was significantly higher than that in the sham-operated group, and the expression of P0.01XIAP in the model group was lower than that in the sham-operated group. The positive cells and caspase3 expression of TUNEL were significantly decreased in the group of acupoints in the electroacupuncture group, and the expression of XIAP was increased in the group of P0.01, which was significantly lower than that in the group of acupoints in the electroacupuncture group. The expression of TUENL and caspase3 was higher in the non-acupoint group than in the control group. Conclusion: electroacupuncture can delay the degeneration of intervertebral disc, which may be related to the regulation of caspase3 and XIAP expression and the inhibition of apoptosis.
