【摘要】：目的:為研究β-胡蘿卜素對(duì)腸上皮細(xì)胞的抗炎作用機(jī)制。方法:選用仔豬空腸上皮(IPEC-J2)細(xì)胞系建立體外細(xì)胞模型,將同一代數(shù)的IPEC-J2細(xì)胞分為4組(空白對(duì)照組、β-胡蘿卜素組、β-胡蘿卜素預(yù)保護(hù)組和脂多糖組)。空白對(duì)照組不做處理,β-胡蘿卜素組和β-胡蘿卜素預(yù)保護(hù)組均進(jìn)行β-胡蘿卜素預(yù)處理,脂多糖(LPS)組和β-胡蘿卜素預(yù)保護(hù)組均采用LPS刺激。MTT法檢測(cè)細(xì)胞活力;Western blot法檢測(cè)Occludin、Claudin4、ZO-1緊密連接蛋白表達(dá)水平。結(jié)果:LPS組IPEC-J2細(xì)胞的緊密連接蛋白表達(dá)水平均低于空白對(duì)照組,且差異有統(tǒng)計(jì)學(xué)意義(P0.05),β-胡蘿卜素預(yù)保護(hù)組緊密連接蛋白表達(dá)水平顯著高于LPS組(P0.05)。結(jié)論:提高緊密連接蛋白表達(dá)水平可能是β-胡蘿卜素對(duì)空腸上皮細(xì)胞的抗炎作用的途徑之一。
[Abstract]:Objective: to study the anti-inflammatory mechanism of 尾 -carotene on intestinal epithelial cells. The IPEC-J2 cells of the same algebra were divided into 4 groups (blank control group, 尾 -carotene group, 尾 -carotene preprotection group and lipopolysaccharide group). The blank control group was not treated, the 尾 -carotene group and 尾 -carotene preprotection group were pretreated with 尾 -carotene. Lipopolysaccharide (LPS) group and 尾 -carotene preprotection group were used to detect the activity of IPEC-J2 cells by LPS stimulation. Western blot assay was used to detect the expression of close-junction protein in IPEC-J2 cells. Results the expression of tight junction protein in IPEC-J2 cells was lower than that in control group. The expression of tight junction protein in 尾 -carotene preprotection group was significantly higher than that in LPS group (P 0.05). Conclusion: increasing the expression level of tight junction protein may be one of the ways of 尾 -carotene anti-inflammatory effect on jejunal epithelial cells.
