【摘要】：目的利用腸道病毒分子定型試驗,對病毒性腦炎患兒的腦脊液標本進行分子型別鑒定。方法提取患兒腦脊液核酸,利用半巢式聚合酶鏈反應(yīng)(Sn-PCR)對腸道病毒VP1區(qū)進行特異性擴增并進行序列測定,通過BLAST比對確定腸道病毒的類型,采用Mega6.0軟件構(gòu)建基因進化樹。結(jié)果 BLAST比對顯示該病毒是腸道病毒中的柯薩奇病毒B5(Coxsackievirus group B5,CVB5)。從進化樹上看,青海分離株04/CVB5/2010/Qinghai/CHN由2009年山東分離株SD2009-067不斷演變而來,而與分離株AH-HK19/CHN/2010/CVB5不在同一進化分支,相距甚遠,與FJLY/2011096h分離株親緣性關(guān)系較近。結(jié)論成功應(yīng)用Sn-PCR對患兒腦脊液中未知腸道病毒進行鑒定,確定腸道病毒類型是CVB5型,為腸道病毒引起的腦炎疾病的防控提供參考。
[Abstract]:Objective to identify the molecular types of cerebrospinal fluid (CSF) samples of children with viral encephalitis by molecular typing test of enterovirus. Methods the nucleic acids of CSF were extracted from children with viral encephalitis. The VP1 region of enterovirus was amplified and sequenced by semi-nested polymerase chain reaction (Sn-PCR). The type of enterovirus was determined by BLAST comparison. Mega6.0 software was used to construct the gene evolutionary tree. Results the BLAST comparison showed that the virus was coxsackievirus B5Coxsackievirus group B5CVB5. From the evolutionary tree, Qinghai isolate 04 / CVB5 / 2010 / Qinghai / Chn evolved continuously from the Shandong isolate SD2009-067 in 2009. However, the relationship between AH-HK19/CHN/2010/CVB5 and AH-HK19/CHN/2010/CVB5 was not the same evolutionary branch, and the genetic relationship was close to that of FJLY/2011096h. Conclusion Sn-PCR was successfully used to identify unknown enterovirus in cerebrospinal fluid of children, and the type of enterovirus was identified as CVB5 type. It provides reference for the prevention and control of encephalitis caused by enterovirus.
