【摘要】：目的:觀察少腹逐瘀湯對子宮內膜異位癥模型大鼠在位內膜增殖、凋亡及血管生成的影響,探討少腹逐瘀湯治療子宮內膜異位癥的機制。方法:58只SD雌性大鼠中隨機選擇10只作為對照組,剩余大鼠采用自體移植法建立子宮內膜異位癥大鼠模型,造模成功的38只大鼠隨機分為模型組及治療組,治療組給予少腹逐瘀湯灌胃,給藥4周。HE染色觀察在位內膜組織形態(tài)及內膜厚度,免疫組化法檢測PCNA表達,TUNEL法檢測細胞凋亡,免疫熒光雙標法檢測CD34、SMA-α表達以評估微血管密度(MVD)及血管成熟指數(shù)(VMI)。結果:模型組在位內膜較對照組、治療組明顯增厚(P0.05),在位內膜凋亡指數(shù)明顯低于對照組及治療組(P0.01),MVD值明顯高于對照組及治療組(P0.05),VMI值明顯低于對照組及治療組(P0.01)。結論:少腹逐瘀湯可以通過誘導子宮內膜異位癥模型大鼠在位內膜間質細胞凋亡來抑制在位內膜增殖,并能夠抑制在位內膜血管生成,促進微血管成熟。
[Abstract]:Objective: to observe the effect of Shaofan Zhuyu decoction on the proliferation, apoptosis and angiogenesis of eutopic endometrium in rats with endometriosis. To explore the mechanism of Shaofan Zhuyu decoction in the treatment of endometriosis. Methods Ten of 58 female SD rats were randomly selected as control group, and the remaining rats were used to establish the model of endometriosis by autologous transplantation. Thirty-eight rats were randomly divided into two groups: model group and treatment group. The rats in the treatment group were treated with Shaofan Zhuyu decoction for 4 weeks. The morphology and thickness of eutopic endometrium were observed by HE staining. The expression of PCNA was detected by Tunel and apoptosis was detected by immunohistochemical method. The expression of CD34 SMA- 偽 was detected by immunofluorescence double labeling method to evaluate the microvessel density (MVD) and vascular maturity index (VMIA). Results: the eutopic endometrium in the model group was higher than that in the control group. The apoptotic index of eutopic endometrium in the treatment group was significantly lower than that in the control group and the treatment group. The MVD value of the treatment group was significantly higher than that of the control group and the treatment group. Conclusion: Shaofangzhuyu decoction can induce uterus by inducing uterus. In order to inhibit the proliferation of eutopic endometrium, the eutopic endometrial stromal cells apoptosis in the rat model of endometriosis. It can inhibit the angiogenesis of eutopic endometrium and promote microvascular maturation.
