IL-12p40與IL-12p70在單病原菌性血流感染小鼠模型中的表達及意義

【摘要】:目的探討白細胞介素-12p40(IL-12p40)和IL-12p70在不同病原菌致小鼠血流感染模型中的動力學變化及意義。方法建立金黃色葡萄球菌、糞腸球菌、大腸埃希菌和肺炎克雷伯菌標準菌株的CD-1(ICR)小鼠血流感染模型并評價;采用蛋白液相芯片檢測技術(shù)測定各實驗組與磷酸緩沖溶液(PBS)的對照組0.5、1、3、6、12、24和48h的IL-12p40和IL-12p70濃度。結(jié)果金黃色葡萄球菌組與對照組相比,IL-12p40的濃度稍升高,而糞腸球菌組、大腸埃希菌組和肺炎克雷伯菌組感染后1h即明顯上升,3h時達峰值,分別為(216.36±50.63)pg/mL、(118.32±9.98)pg/mL和(129.19±27.11)pg/mL。感染12h后仍維持較高水平,與對照組相比差異有統(tǒng)計學意義(P0.05);金黃色葡萄球菌組IL-12p70濃度在感染后6h時明顯升高且達峰值,為(389.31±118.07)pg/ml;糞腸球菌組IL-12p70濃度在感染后3h明顯升高,與對照組相比差異有統(tǒng)計學意義(P0.05),大腸埃希菌組與肺炎克雷伯菌組IL-12p70濃度于感染后3h達峰值,分別為(42.20±3.54)pg/mL和(62.80±8.37)pg/mL但升高幅度顯著低于金黃色葡萄球菌組和糞腸球菌組(P0.05)。結(jié)論 IL-12p40和IL-12p70在四組單病原菌性血流感染模型早期均明顯升高,感染金黃色葡萄球菌或糞腸球菌后IL-12p70濃度的升高程度明顯高于大腸埃希菌或肺炎克雷伯菌感染。聯(lián)合檢測IL-12p40和IL-12p70等指標對區(qū)分革蘭陰陽性菌感染有一定價值。
[Abstract]:Objective to investigate the dynamic changes and significance of interleukin-12p40 IL-12p40 (IL-12p40) and IL-12p70 in blood stream infection model of mice induced by different pathogens. Methods Staphylococcus aureus and Enterococcus faecalis were established. Model of blood stream infection in mice with standard strains of Escherichia coli and Klebsiella pneumoniae; The concentrations of IL-12p40 and IL-12p70 in each experimental group and control group with phosphoric acid buffer solution were measured by protein liquid chip assay at 24 and 48 hours. Results the concentration of IL-12p40 in Staphylococcus aureus group was slightly higher than that in control group, while that in Enterococcus faecalis group was higher than that in Staphylococcus aureus group. In Escherichia coli group and Klebsiella pneumoniae group, the peak value was 216.36 鹵50.63 g / mL and 129.19 鹵27.11 渭 g / mL respectively at 3 h after infection. Compared with the control group, the concentration of IL-12p70 in Staphylococcus aureus group increased significantly and reached its peak at 6 h after infection, which was 389.31 鹵118.07 mg / ml, while the concentration of IL-12p70 in Enterococcus faecalis group increased significantly at 3 h after infection. The concentration of IL-12p70 in Escherichia coli group and Klebsiella pneumoniae group reached the peak at 3 h after infection. It was 42.20 鹵3.54 ng / mL and 62.80 鹵8.37 ng / mL, respectively, but the increase was significantly lower than that in Staphylococcus aureus group and Enterococcus faecalis group (P 0.05). Conclusion IL-12p40 and IL-12p70 increased significantly in the early stage of single pathogen blood stream infection in the four groups. The concentration of IL-12p70 after infection with Staphylococcus aureus or Enterococcus faecalis was significantly higher than that with Escherichia coli or Klebsiella pneumoniae, and the combined detection of IL-12p40 and IL-12p70 had some value in differentiating Gram-positive infection.
